作者以U_(14)(小鼠宫颈癌第14号)细胞为抗原,免疫BALB/C小鼠,取其脾细胞与NS—1小鼠骨髓瘤细胞在PEG(聚乙二醇)作用下进行细胞融合。所形成的淋巴细胞杂交瘤细胞能生长于一种选择性培养液—HAT培养液中。对于具有分泌特异抗体的杂交瘤细胞,借助其上清液的免疫荧光试验加以选择。将间接膜免疫荧光试验阳性的杂交瘤细胞用有限稀释法进行克隆和扩大培养,得到1株专一分泌抗U_(14)单克隆抗体的淋巴细胞杂交瘤,定名为AU_(14-1)。杂交瘤细胞正继续在培养中。
In this study, BALB / C mice were immunized with U14 (mouse cervical cancer cell No. 14) as antigen and their spleen cells and NS-1 mouse myeloma cells were treated with PEG (polyethylene glycol) Cell fusion. The resulting lymphocyte hybridoma cells can grow in a selective medium-HAT medium. For hybridoma cells with secreted specific antibodies, their supernatants were selected by immunofluorescence assay. The indirect hybridization immunofluorescence assay hybridoma cells were cloned and expanded by limiting dilution method to obtain a monoclonal antibody secreting anti-U14 monoclonal antibody (named as AU_ (14-1)). Hybridoma cells are continuing to be cultured.
