目的:观察雷公藤多苷(Tripterygium Glycosides,TG)对大鼠卵巢功能耗损的影响,并探讨菟丝子提取物对其卵巢功能的保护和修复作用。方法:将SD大鼠50只随机分为A、B1、B2、C1、C2共5组,每组10只。A为空白对照组,每天0.1m L 0.9%氯化钠注射液灌胃;B1、B2为TG给药组,于每天8∶0、20∶00分别灌胃TG 50mg/kg/d,连续给药14d;C1、C2为菟丝子提取物给药组,每天灌胃菟丝子提取物895.69mg/kg,给药2次,上午下午各1次,连续3d。HE染色观察卵巢组织形态,放射免疫法检测血清雌二醇(E_2)、促卵泡刺激素(FSH)、促黄体生成激素(LH)水平。结果:与A组比较,B1、B2组动情周期延长为7~8d/次,卵巢萎缩,结构受损。E_2、FSH、LH水平均无明显改变;而C1、C2组动情周期延长为7~8d/次,偶为5d/次,卵巢结构病变减轻,E_2、FSH及LH水平均无明显改变。结论:TG可致大鼠卵巢功能损害,晚上给药较白天给药损害较轻,菟丝子提取物对其生殖系统损害有明显改善作用。
OBJECTIVE: To observe the effect of Tripterygium Glycosides (TG) on ovarian dysfunction in rats and to explore the protective and repair effects of dodder extract on ovarian function. Methods: 50 SD rats were randomly divided into A, B1, B2, C1, C2, a total of 5 groups of 10. A is a blank control group, 0.1 ml 0.9% sodium chloride injection daily; B1, B2 for the TG administration group, respectively, at 8:020:00 daily gavage TG 50mg / kg / d, continuous Drug 14d; C1, C2 dodder extract administration group, daily dipped dodder extract 895.69mg / kg, administered twice in the afternoon 1 each for 3 consecutive days. The morphological changes of ovarian tissue were observed by HE staining. Serum estradiol (E2), follicle stimulating hormone (FSH) and luteinizing hormone (LH) were measured by radioimmunoassay. Results: Compared with group A, the estrous cycle in groups B1 and B2 was prolonged to 7-8 days, and the ovary atrophy and the structure was damaged. E_2, FSH and LH did not change significantly. The estrous cycle in groups C1 and C2 was prolonged to 7-8 days / even for 5 days. The ovarian lesions lessened and the levels of E2, FSH and LH were not changed. CONCLUSION: TG can cause ovarian dysfunction in rats, which is less damaging than administration in the evening, and that Dodder extract has a significant effect on the reproductive system damage.
