目的:观察龟鹿二仙胶汤含药血清干预软骨细胞表型化骨髓质基质干细胞(BMSCs)凋亡的影响。方法:抽取兔骨髓液,经体外分离、纯化、培养获得免BMSCs,将龟鹿二仙胶汤含药血清诱导出的软骨细胞表型化BMSCs,用IL-1β诱导凋亡,分别于24h、48h、72h后,采用透射电子显微镜、免疫荧光组织化学、流式细胞仪等实验技术,研究龟鹿二仙胶汤对软骨表型化BMSCs凋亡的影响。结果:龟鹿二仙胶汤能减少软骨表型化BMSCs凋亡的数量及程度,降低细胞凋亡率。结论:龟鹿二仙胶汤可以明显抑制或逆转软骨表型化BMSCs凋亡,能显著延缓软骨表型化BMSCs的凋亡。
Objective: To observe the effect of Guilu Erxian Decoction serum on the apoptosis of chondrocyte phenotype mesenchymal stem cells (BMSCs). METHODS: Rabbit bone marrow fluid was extracted and isolated, purified and cultured in vitro to obtain BMSCs-free. The chondrocytes induced by Guilu Erxian Decoction serum were phenotype BMSCs and apoptosis was induced by IL-1β at 24 h. After 48h and 72h, the effect of Guilu Erxianjiao decoction on apoptosis of cartilage phenotyped BMSCs was studied by transmission electron microscopy, immunofluorescence histochemistry and flow cytometry. RESULTS: Guilu Erxiantang Decoction can reduce the number and degree of apoptosis of cartilage phenotype BMSCs and reduce the rate of apoptosis. Conclusion: Guilu Erxian Decoction can obviously inhibit or reverse the apoptosis of cartilage phenotyped BMSCs, and can significantly delay the apoptosis of cartilage phenotyped BMSCs.
