目的 :探讨他莫昔芬对雌二醇抑制大鼠颈总动脉球囊损伤后内膜增生的影响。方法 :8~ 10周龄 SD大鼠 ,分为雄性组和雌性组 ,每组 2 1只 ,各组又平分为 3个亚组 :去势对照组 ,雌二醇组 ,他莫昔芬组。应用 F2 .0 经皮腔内冠状动脉成形术 ( PTCA)球囊导管损伤血管内膜。2周后处死动物 ,取出左、右颈总动脉作切片观察。结果 :雄性组 :内膜面积、内膜面积 /肌层面积值 ,雌二醇组均小于去势对照组 (均 P <0 .0 5 ) ;他莫昔芬组与去势对照组无显著性差异 (均 P >0 .0 5 )。雌性组 :内膜面积、内膜面积 /肌层面积值 ,雌二醇组均小于去势对照组 (均 P <0 .0 5 ) ,而他莫昔芬组与去势对照组相比亦无统计学差异 (均 P >0 .0 5 )。结论 :雌二醇对去势雄性、雌性大鼠颈总动脉损伤后均有抑制内膜增生作用 ;他莫昔芬拮抗雌二醇的作用 ,提示雌二醇通过受体抑制内膜增生
Objective: To investigate the effect of tamoxifen on the intimal hyperplasia after estradiol inhibits the common carotid artery balloon injury in rats. Methods: Sprague-Dawley (SD) rats aged 8-10 weeks were divided into male and female group, with 21 in each group. Each group was equally divided into 3 subgroups: castration control group, estradiol group, tamoxifen group . Percutaneous transluminal coronary angioplasty (PTCA) balloon catheter was used to injure the vascular intima. After 2 weeks, the animals were sacrificed and the left and right common carotid arteries were sectioned for observation. Results: In the male group, the intimal area, intima-area / myometrial area value were significantly lower in the estradiol group than those in the castration control group (all P <0.05), but not in the tamoxifen group and the castrated control group Sex differences (all P> 0.05). Female group: intimal area, intima area / myometrial area value, estradiol group were less than castration control group (all P <0.05), while tamoxifen group compared with castration control group No statistical difference (all P> 0.05). CONCLUSION: Estradiol inhibits intimal hyperplasia after carotid artery injury in castrated male and female rats. Tamoxifen antagonizes the effect of estradiol, suggesting that estradiol inhibits intimal hyperplasia via receptor
