目的:观察麻荆止咳颗粒对咳嗽变异性哮喘豚鼠模型肺组织及外周血中Eotaxin表达的影响。方法:采用卵白蛋白致敏法制备咳嗽变异性哮喘的豚鼠模型。用酶联免疫吸附测定(ELISA)法测定各组豚鼠外周血中Eotaxin含量,用逆转录PCR技术测定各组豚鼠肺组织中Eotaxin的表达。结果:各组肺组织中Eotaxin的含量显示,空白对照组最低,模型组明显升高(P<0.01),经低、中、高剂量麻荆止咳颗粒干预后Eotaxin含量明显降低,且有统计学意义(P<0.01)。在豚鼠外周血中,随着药物剂量的增大,Eotaxin含量逐步降低;高剂量组与模型组比较差异有统计学意义(P<0.05),说明高剂量组能降低造模豚鼠血清中Eotaxin的含量。结论:麻荆止咳颗粒减轻哮喘气道炎症与其抑制Eotaxin表达有关。
Objective: To observe the effect of Ma Jing Zhike Granule on the expression of Eotaxin in the lung tissue and peripheral blood of the cough variant asthmatic guinea pig model. Methods: Guinea pig model of cough variant asthma was made by ovalbumin sensitization. The levels of Eotaxin in peripheral blood of guinea pigs in each group were determined by enzyme-linked immunosorbent assay (ELISA), and the expression of Eotaxin in lungs of guinea pigs were determined by reverse transcription polymerase chain reaction (RT-PCR). Results: The content of Eotaxin in lung tissue of each group showed that the content of Eotaxin in the blank control group was significantly lower than that in the untreated group (P <0.01) Significance (P <0.01). In guinea pig peripheral blood, the content of Eotaxin gradually decreased with the increase of drug dose; the difference between high dose group and model group was statistically significant (P <0.05), indicating that high dose group can reduce Eotaxin content. Conclusion: Ma Jing Zhike Granule can reduce the airway inflammation in asthma and its inhibition of Eotaxin expression.
