目的建立HPLC法测定复方葛根片中隐丹参酮和丹参酮ⅡA含量方法,以便更好的控制复方葛根片质量。方法采用HPLC法,色谱柱:SHMADZU VP-ODS柱(250 mm×4.6 mm,5μm);流动相:乙腈(A)-0.05%-磷酸水溶液(B)为流动相进行梯度洗脱:0~15 min,10%~50%A;15~25 min,50%~80%A;25~30 min,80%~90%A;检测波长:270 nm;柱温:30℃;流速:1.0 ml·min~(-1)。结果隐丹参酮在0.10~0.25μg范围线性关系良好(r=1),丹参酮ⅡA在0.11~0.275μg范围线性关系良好(r=0.999);平均回收率分别是99.83%、98.98%,RSD值分别是1.08%、0.81%。结论本方法简便可行,重复性好,可用于该制剂的质量检测。
OBJECTIVE To establish a HPLC method for the determination of cryptotanshinone and tanshinone Ⅱ A in Pueraria lobata in order to better control the quality of Pueraria lobata. Methods The mobile phase consisted of acetonitrile (A) -0.05% - phosphoric acid solution (B) with gradient elution gradient of 0-15 on a SHMADZU VP-ODS column (250 mm × 4.6 mm, min, 10% -50% A, 15-25 min, 50-80% A, 25-30 min, 80% -90% A, detection wavelength: 270 nm, column temperature: min ~ (-1). Results The results showed that there was a good linear relationship between cryptotanshinone in the range of 0.10-0.25 μg (r = 1) and the linear range of 0.11-0.275 μg (r = 0.999) in the tanshinone Ⅱ A group. The average recoveries were 99.83% and 98.98% 1.08%, 0.81%. Conclusion This method is simple and feasible, reproducible, and can be used for the quality testing of the preparation.
