目的:研究脉络宁注射液(Mailuoning injection,MLN)对手术结扎血瘀模型小鼠的保护作用。方法:取雄性KM小鼠50只,随机分为空白对照组,模型对照组,脉络宁低、中、高剂量组(3.75、7.5、15 mg/kg),腹腔注射给药,给药14 d,每天2次。末次给药2 h后通过手术结扎下腔静脉的方法造成血瘀模型,手术5 h后,用各组小鼠眼眶静脉血检测凝血时间、白介素-6(IL-6)、肿瘤坏死因子α(TNFα)和组织因子(tissue factor,TF)的含量,术后6 h,麻醉解剖小鼠,取出下腔静脉作血管长度重量比值的检测和蛋白免疫印迹检测TF表达含量,取出肝、脾、胸腺作脏器系数和肝、脾病理学检测。结果:通过脉络宁注射液治疗后,与模型对照组相比较,脉络宁各剂量组的凝血时间增长(P<0.05)和IL-6、TNFα、TF含量减少(P<0.05或P<0.01);脉络宁高剂量组的肝和脾的脏器系数减少(P<0.05),胸腺的脏器系数都增加(P<0.01);脉络宁高剂量组的血管重量长度比和TF表达都减少(P<0.05),脉络宁各剂量组肝脾的血瘀症状得到改善。结论:脉络宁注射液对手术结扎血瘀模型小鼠有一定化瘀作用。
Objective: To study the protective effect of Mailuoning injection (MLN) on blood stasis model in mice. Methods: Fifty male KM mice were randomly divided into blank control group, model control group, Mailuoning low, medium and high dose groups (3.75, 7.5, 15 mg / kg), intraperitoneal injection for 14 days , 2 times a day. The blood stasis model was established by surgical ligation of the inferior vena cava 2 h after the last administration. After 5 h of operation, the orbital venous blood of each group was used to detect the clotting time, interleukin-6 (IL-6), tumor necrosis factor α (TNFα) and tissue factor (TF). Six hours after operation, the rats were sacrificed and the inferior vena cava was taken out to measure the ratio of vascular length and weight. TF expression was detected by Western blotting, and the liver, spleen and thymus Organ coefficient and liver, spleen pathological examination. Results: Compared with the model control group, the contents of IL-6, TNFα and TF were decreased (P <0.05 or P <0.01) (P <0.05). The organ coefficient of the thymus increased (P <0.01), while that of the Mailuoning high dose group decreased (P <0.05) P <0.05), Mailuoning dose of liver and spleen of blood stasis symptoms were improved. Conclusion: Mailuoning injection on blood stasis ligation model mice have some blood stasis.
