应用核团微量注射、放射免疫测定(RIA)和高压液相(HPLC)观察了刺激视上核(SON)对蓝斑(LC)灌流液中催产素(OT)、精氨酸加压素(AVP)、去甲肾上腺素(NE)和5-羟色胺(5-HT)的含量的变化以及蓝斑注射OT和AVP的拮抗剂对痛阈(PT)的影响。结果表明:刺激SON后30到90min,LC灌流液中OT的含量,30minAVP的含量,60min5-HT的含量明显增加,而NE的含量在30和60min时明显降低。注射AVP的V1拮抗剂到LC对注射L-谷氨酸到SON引起的镇痛作用无影响,V2拮抗剂可部分抑制和OT拮抗剂可翻转刺激SON引起的镇痛作用。以上结果提示刺激SON引起的镇痛作用可能是由于SON释放的OT通过作用于LC内的OT和V2受体使LC内的5-HT增加和NE降低而产生的。
The effect of supraoptic nucleus (SON) on oxytocin (OT), arginine vasopressin (AVP), norepinephrine (NE) and serotonin (5-HT) levels in rats and the effect of locus coeruleus OT and AVP antagonists on pain threshold (PT). The results showed that the contents of OT, 30 min and 60 min 5-HT in LC perfusate increased significantly from 30 to 90 min after stimulating SON, while the content of NE significantly decreased at 30 and 60 min. The injection of V1 antagonist of AVP into LC had no effect on the analgesic effect caused by injection of L-glutamic acid to SON, while V2 antagonist partially inhibited the analgesic effect of OT antagonist reversible stimulation of SON. The above results suggest that the stimulation of the analgesic effect caused by SON may be due to the OT released by SON resulting from increased 5-HT and NE reduction in the LC by acting on the OT and V2 receptors within the LC.
