目的:观察桃核承气汤及其拆方组对蓄血症大鼠模型“瘀热”相关指标的影响。方法:间隔24 h两次大鼠尾静脉注射内毒素脂多糖(lipopolysaccharide,LPS)50μg.kg-1造模。第1次尾静脉注射后0.5,12 h,第2次尾静脉注射后0.5 h灌胃给药。给药组剂量全方组10 g.kg-1,调胃组5.7 g.kg-1,调桃组8.6 g.kg-1,调桂组7.1 g.kg-1,桃桂组4.3 g.kg-1。模型对照组,空白对照组给相应剂量生理盐水。末次注射后2 h取血,检测血清中组织纤溶酶原激活物(tissue type plasminogenactivator,t-PA),纤溶酶原激活物抑制剂-1(plasminogen activator inhibitor,PAI-1),超氧化物歧化酶(superoxide dismutase,SOD),丙二醛(malondialdehyde,MDA),一氧化氮(nitric oxide,NO)及血浆中内皮素-1(endothelin-1,ET-1),6-酮-前列腺素-1α(6-keto-prostaglandin F1α,6-keto-F1α),血栓烷素B2(thromboxane B2,TXB2)。结果:全方组,调胃组,调桂组,调桃组能上调SOD(P<0.01),下调MDA(P<0.01)。全方组,调胃组,调桂组能下调PAI-1及t-PA/PAI-1(P<0.01)。调胃组,调桂组还能上调t-PA(P<0.01)。桃桂组对SOD,MDA及PAI-1干预作用小,与全方组比较有差异(P<0.01)。各给药组均能升高大鼠血浆中TXB2,降低6-Keto-PGF1α/TXB2,降低NO。结论:桃核承气汤对“瘀热”指标均有积极的干预作用。各拆方组中,调胃组处于核心地位,对“瘀热”指标作用均比较好。调桂组对反映“瘀”的指标干预作用好于全方组。桃桂组在调解“瘀热”指标中表现均最差。
Objective: To observe the effect of Taohe Chengqi Decoction and its disassembled prescriptions on the related indexes of “stasis” in rat model of hematemia. Methods: Rats were injected intravenously with lipopolysaccharide (LPS) 50μg · kg-1 twice a day for 24 h. 0.5,12 h after the first tail vein injection and 0.5 h after the second tail vein injection. The dosage of the treatment group was 10 g.kg-1 for the whole-dose group, 5.7 g.kg-1 for the modified stomach group, 8.6 g.kg-1 for the turtle group, 7.1 g.kg-1 for the tui-gui group and 4.3 g for the Tao-gui group. kg-1. The blood samples were collected 2 h after the last injection, and the serum levels of tissue-type plasminogen activator (t-PA), plasminogen activator inhibitor (PAI-1) Superoxide dismutase (SOD), malondialdehyde (MDA), nitric oxide (NO) and plasma endothelin-1 (ET-1) 6-keto-prostaglandin F1α, 6-keto-F1α and thromboxane B2 (TXB2). Results: The whole group, adjusting stomach group, adjusting Gui group and Toutao group could up-regulate SOD (P <0.01) and down-regulate MDA (P <0.01). The whole group, tune the stomach group, tune Gui group can reduce PAI-1 and t-PA / PAI-1 (P <0.01). Tiao Wei group, tune Gui group also increased t-PA (P <0.01). Tao Gui group intervention of SOD, MDA and PAI-1 small, compared with the whole group were significantly different (P <0.01). Each administration group could increase TXB2 in plasma, decrease 6-Keto-PGF1α / TXB2 and decrease NO. Conclusion: Taohe Chengqi Decoction has a positive intervention on “stasis” index. In each split group, tune the stomach group is at the core of the “stasis ” indicators are better. Gui Gui group to reflect the “stasis ” index intervention better than the whole group. Tao Gui group mediation “stasis ” indicators were the worst performance.
