目的了解微生物实验室压力蒸汽灭菌效果状况,以提高和保证压力蒸汽灭菌质量。方法采用工艺监测和生物指示剂监测方法,对实验室压力蒸汽灭菌质量进行了验证和监控。结果经对所选择的压力蒸汽灭菌器的灭菌工艺验证,装载容量控制在70%条件下,对盛装于500ml三角烧瓶培养基的灭菌工艺确定为121℃20min。用留点温度计进行252次灭菌温度监测,灭菌温度合格率为100%,化学指示剂监测合格率为98.0%。在设定灭菌温度121℃,作用时间20min,用嗜热脂肪芽孢杆菌芽孢进行15次灭菌效果监测,合格率为100%。结论经过工艺验证,所确定的灭菌条件,可确保压力蒸汽灭菌质量。
Objective To understand the effect of pressure steam sterilization in microbiology laboratory to improve and ensure the quality of pressure steam sterilization. Methods Process monitoring and biological indicator monitoring methods were used to verify and monitor the quality of laboratory steam sterilization. Results The sterilization process of the selected pressure steam sterilizer verified that the sterilization process at 500 ml Erlenmeyer flask medium was determined to be 121 ° C for 20 minutes under the condition that the loading capacity was controlled at 70%. 252 stay sterilization temperature monitoring points with a retention point, the sterilization temperature pass rate of 100%, the chemical indicator monitoring pass rate was 98.0%.
