目的:观察CD40-RNAi抑制CD40-CD40L相互作用对ApoE-/-小鼠血小板功能的影响。方法:将ApoE-/-小鼠分为anti-CD40L抗体组、CD40-RNAi组及对照组,应用流式细胞术分别检测血小板膜上表达P-选择素(CD62P)水平及血小板-单核细胞聚集率。结果:ApoE-/-小鼠anti-CD40L抗体组(15.6±3.3,MFI)及CD40-RNAi组(10.7±2.7,MFI)血小板膜表达CD62P水平明显较对照组低(21.8±3.6,MFI);同时发现血小板-单核细胞聚集率anti-CD40L抗体组(11.2±2.3)%及CD40-RNAi组(6.2±2.7)%较对照组低(19.3±2.6)%。ApoE-/-小鼠CD40-RNAi干预前血小板表达CD40水平(67.3±11.3,MFI)明显高于干预后(6.5±1.9,MFI)。结论:CD40-RNAi通过阻断CD40-CD40L相互作用抑制血小板活化功能。
Objective: To observe the effect of CD40-RNAi on CD40-CD40L interaction on platelet function in ApoE - / - mice. Methods: ApoE - / - mice were divided into anti-CD40L antibody group, CD40-RNAi group and control group. The level of CD40 on platelet (67.3 ± 11.3, MFI) before CD40-RNAi intervention in ApoE - / - mice was significantly higher than that after intervention (6.5 ± 1.9, MFI). Conclusion: CD40-RNAi inhibits platelet activation by blocking the interaction of CD40-CD40L.
