目的:研究肿瘤坏死因子相关凋亡诱导配体(TRAIL)受体在胰腺癌中的表达及意义。方法:应用半定量RT-PCR,检测TRAILR mRNA在胰腺癌组织,正常胰腺组织及胰腺癌细胞系ASPC-1、Can-pan-2中的表达。结果:死亡受体DR4、DR5在所有胰腺癌组织、正常胰腺组织及胰腺癌细胞系中均有表达,诱骗受体DcR1、DcR2在所有正常胰腺组织及细胞系中均有表达。死亡受体DR4、DR5在胰腺癌组织中有较高的表达,而在正常胰腺组织中呈中低水平表达(P<0.01)。胰腺癌细胞系中死亡受体DR4、DR5呈高水平表达,而诱骗受体DcR1、DcR2仅呈中低水平表达。结论:TRAIL受体在胰腺癌普遍表达,并存在受体类型的表达差异;死亡受体在胰腺癌中高表达,可能在TRAIL诱导胰腺癌细胞凋亡的机制中发挥重要的作用。
Objective: To study the expression of tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) receptor in pancreatic cancer and its significance. Methods: The expression of TRAILR mRNA in pancreatic cancer tissues, normal pancreatic tissues and pancreatic cancer cell lines ASPC-1 and Can-pan-2 was detected by semi-quantitative RT-PCR. Results: The death receptors DR4 and DR5 were expressed in all pancreatic cancer tissues, normal pancreatic tissues and pancreatic cancer cell lines. The decoy receptors DcR1 and DcR2 were expressed in all normal pancreatic tissues and cell lines. DR4 and DR5, the death receptors, were highly expressed in pancreatic cancer tissues and moderately and lowly expressed in normal pancreatic tissues (P <0.01). The DR4 and DR5 death receptors in pancreatic cancer cell lines were highly expressed, while the decoy receptors DcR1 and DcR2 were only expressed at low and middle levels. CONCLUSION: TRAIL receptor is commonly expressed in pancreatic cancer, and there is a difference in the expression of receptor types. Death receptor is highly expressed in pancreatic cancer and may play an important role in the mechanism of TRAIL-induced apoptosis in pancreatic cancer cells.
