目的:通过研究自发性高血压大鼠(SHR)外周血淋巴细胞钙激活钾通道KCa3.1表达的变化,为高血压病淋巴细胞激活提供证据。方法:使用密度离心法分离SHR和Wistar大鼠外周血淋巴细胞,运用实时荧光定量PCR和Western blot技术检测SHR和Wistar大鼠淋巴细胞KCa3.1基因和蛋白的表达。结果:(1)SHR组KCa3.1 mRNA表达水平高于对照组(P<0.05);(2)SHR组KCa3.1蛋白表达水平高于对照组(P<0.05)。结论:SHR淋巴细胞KCa3.1表达增多,提示KCa通道在调节高血压病淋巴细胞激活中起着重要的作用。
Objective: To investigate the changes of KCa3.1 expression in the calcium-activated potassium channel of peripheral blood lymphocytes in spontaneously hypertensive rats (SHRs) and provide evidence for the activation of lymphocytes in hypertensive patients. Methods: Peripheral blood lymphocytes of SHR and Wistar rats were isolated by density centrifugation. The expression of KCa3.1 gene and protein in lymphocytes of SHR and Wistar rats were detected by real-time fluorescence quantitative PCR and Western blot. Results: (1) The expression of KCa3.1 mRNA in SHR group was higher than that in control group (P <0.05). (2) The expression of KCa3.1 protein in SHR group was higher than that in control group (P <0.05). Conclusion: The increased expression of KCa3.1 in SHR lymphocytes suggests that KCa channel plays an important role in the regulation of hypertensive lymphocyte activation.
