目的:建立中药五味子中7个成分在血浆药物测定方法,探讨它们在大鼠体内药代动力学情况。方法:采用交叉给药设计,应用高效液相色谱法测定Wistar大鼠单次单剂量灌胃五味子醇提物(0.5 g药材/kg)后的血液中五味子醇甲、戈米辛D、五味子醇乙、五味子酯甲、五味子甲素、五味子乙素、五味子丙素的浓度,利用DAS 2.0软件计算药动学参数。结果:大鼠血浆中7种木脂素成分的校正曲线线性关系良好,日内、日间精密度和准确度符合要求,提取回收率77.2%~90.4%。Wistar大鼠口服灌胃五味子醇提物后,7个成分在大鼠体内的药-时数据拟合后均为二室开放模型。结论:该方法简便、快速、灵敏、稳定,适用于五味子提取物的体内分析,为其进一步研究奠定基础。
OBJECTIVE: To establish a method for the determination of seven components in Chinese medicinal herb Schisandrae by plasma pharmacokinetics and investigate their pharmacokinetics in rats. Methods: The cross-administration design was used to determine the content of Schisandrin A, Gomisin D, Schizandrol in Wistar rats after single single-dose oral administration of Schisandra chinensis (0.5 g medicinal material / kg) by high performance liquid chromatography B, Schisandra ester A, Schisandra A, Schisandrin B, Schisandra B concentration, the pharmacokinetic parameters were calculated using DAS 2.0 software. Results: The calibration curve of seven lignans in plasma of rats showed a good linearity. The precision and accuracy of intra-day and inter-day complied with the requirements. The recovery rates ranged from 77.2% to 90.4%. Wistar rats oral administration of alcohol extract of Schisandra, seven components in the rat drug-time data fitting two-compartment open model. Conclusion: The method is simple, rapid, sensitive and stable and suitable for the in vivo analysis of Schisandra chinensis extract, which lays the foundation for further research.
