目的对天津市2010年的手足口病(HFMD)病例进行病原学分析,并对引起该市HFMD流行的肠道EV71型和柯萨奇病毒A组16型(Cox A16)进行分子生物学特征分析。方法利用Real-time PCR检测HFMD病例标本,用人横纹肌肉瘤(RD)细胞对Real-time PCR检测阳性的部分病例标本进行病毒分离,用RT-PCR法扩增VP1全基因,使用生物学软件进行序列和系统发生分析。结果共检测HFMD病例1766例,肠道病毒总检测阳性率为71.52%(1263/1766),其中EV71占43.94%,Cox A16占41.65%,其他EV占14.41%。15株EV71分离株与C4亚型的C4a分支代表株具有最高的核苷酸序列同源性,均归属于C4a分支;9株Cox A16分离株与B1亚型的代表株具有最高的核苷酸序列同源性,其中8株属于B1b分支,1株属于B1a分支。结论导致天津市2010年HFMD流行的EV71流行株为C4亚型中的C4a病毒株,Cox A16流行株为B1亚型的B1b病毒株。
Objective To analyze the etiology of hand, foot and mouth disease (HFMD) cases in Tianjin in 2010 and analyze the molecular biological characteristics of enteric EV71 and Cox A16 type 16 . Methods Real-time PCR was used to detect HFMD cases. Rhabdomyosarcoma (RD) cells were used to isolate the virus from some cases detected by Real-time PCR. The whole VP1 gene was amplified by RT-PCR and sequenced using biological software And system analysis. Results A total of 1766 cases of HFMD were detected. The positive rate of total enterovirus detection was 71.52% (1263/1766), of which, EV71 accounted for 43.94%, Cox A16 accounted for 41.65% and other EVs accounted for 14.41%. 15 strains of EV71 isolates had the highest nucleotide sequence homology with the C4a subtype of C4 subtype, which belonged to C4a branch. The 9 strains of Cox A16 isolates and B1 subtype had the highest nucleotide Among them, 8 belonged to B1b and 1 belonged to B1a. Conclusions The EV71 epidemic strain that caused HFMD epidemic in Tianjin in 2010 was C4a strain in C4 subtype and the Cox A16 epidemic strain was B1 subtype B1b strain.
