目的:观察长期使用葛根素对去卵巢大鼠阴道壁结构的影响,为中西药葛根素与雌激素组合治疗Ⅰ型骨质疏松症提供实验依据。方法:5月龄健康雌性大白鼠40只,分成5个实验组(n=8只),除假手术组外均建立去卵巢动物模型。①假手术组(Sham);②去卵巢模型组(OVX);③葛根素-50组(Pr-50),皮下注射葛根素50 mg/kg,1次/天;④雌二醇-200组(E2-200),皮下注射雌二醇200μg/kg,2次/周;⑤葛根素-25+雌二醇-100组(Pr-25+E2-100),皮下注射葛根素25 mg/kg,1次/天和雌二醇100μg/kg,2次/周。在第20周,取大鼠血清和阴道壁,观察血清雌二醇(E2)水平和阴道壁结构的变化。结果:OVX组E2水平、阴道上皮及肌层厚度明显低于其他各组(P﹤0.05);E2-200组E2水平、阴道上皮及肌层厚度明显高于其他各组(P﹤0.05);Pr-25+E2-100组E2水平较Pr-50组高(P﹤0.05),但两组间阴道上皮及肌层厚度差异无统计学意义(P﹥0.05)。结论:较大剂量雌二醇可明显促进去卵巢大鼠阴道上皮和肌层增生;小剂量的雌二醇与葛根素联合、较大剂量的葛根素对去卵巢大鼠阴道上皮和肌层增生作用不明显。
OBJECTIVE: To observe the effect of long-term use of puerarin on vaginal wall structure in ovariectomized rats and provide experimental evidence for the combination of puerarin with estrogen and traditional Chinese medicine and western medicine in the treatment of type Ⅰ osteoporosis. Methods: Forty healthy 5-month-old female rats were divided into 5 experimental groups (n = 8), and ovariectomized animals were established except the sham operation group. ① Sham group; ② ovariectomized group (OVX); ③ Puerarin-50 group (Pr-50), subcutaneous injection of puerarin 50 mg / kg, (E2-200), subcutaneous injection of estradiol 200μg / kg, twice a week; ⑤ Puerarin -25 + estradiol -100 group (Pr-25 + E2-100), subcutaneous injection of puerarin 25 mg / kg , Once daily and estradiol 100 μg / kg twice daily. At week 20, rat serum and vaginal wall were taken for observation of the changes of serum estradiol (E2) level and vaginal wall structure. Results: The level of E2, the thickness of vaginal epithelium and muscular layer in OVX group were significantly lower than those in other groups (P <0.05). The E2 level, the thickness of vaginal epithelium and muscular layer in E2 - 200 group were significantly higher than those in other groups (P <0.05). The E2 level in Pr-25 + E2-100 group was higher than that in Pr-50 group (P <0.05), but there was no significant difference between the two groups in the thickness of vaginal epithelium and muscular layer (P> 0.05). Conclusion: Higher doses of estradiol can significantly promote vaginal epithelial and myometrial hyperplasia in ovariectomized rats. Low doses of estradiol and puerarin combined with higher doses of puerarin on vaginal epithelial and myometrial hyperplasia in ovariectomized rats The role is not obvious.
