目的探讨白介素-13(IL-13)对正常人表皮成纤维细胞(NHDF)产生炎症因子的影响。方法使用不同浓度IL-13(0.1、1、10ng/ml)刺激体外培养的NHDF,应用ELISA方法检测IL-13刺激NHDF24h后,白介素-6(IL-6),白介素-8(IL-8),巨噬细胞趋化蛋白-1(MCP-1)和嗜酸性粒细胞趋化因子(Eotaxin)的蛋白表达水平。结果0.1ng/mlIL-13刺激下,IL-6、MCP-1和Eotaxin的蛋白表达水平分别为(727.33±39.88)pg/ml、(414.67±13.01)pg/ml、(124.67±8.67)pg/ml;1ng/mlIL-13刺激下,分别为(1222.50±49.95)pg/ml、(705.33±25.15)pg/ml、(205.33±16.59)pg/ml;10ng/mlIL-13刺激下,分别为(1502.83±5.31)pg/ml、(1329.17±56.08)pg/ml、(457.50±15.32)pg/ml,均明显高于对照组[(283.50±15.86)pg/ml、(205.17±3.76)pg/ml、(13.50±5.01)pg/ml,P<0.05或0.01],并具有浓度依赖性。IL-8蛋白表达水平与对照组无显著性差异(P>0.05)。结论IL-13可刺激NHDF产生IL-6、MCP-1和Eotaxin,这些炎症因子的大量表达可能参与了IL-13在皮肤组织纤维化过程中的病理性作用。
Objective To investigate the effect of interleukin-13 (IL-13) on the production of inflammatory cytokines by normal human epidermal fibroblasts (NHDFs). Methods The NHDF cultured in vitro was stimulated with different concentrations of IL-13 (0.1, 1, 10ng / ml). IL-6, IL-8 and IL- , Macrophage chemotactic protein-1 (MCP-1) and eotaxin. Results The protein expression levels of IL-6, MCP-1 and Eotaxin were (727.33 ± 39.88) pg / ml, (414.67 ± 13.01) pg / ml and (124.67 ± 8.67) pg / ml and (205.33 ± 16.59) pg / ml and 10ng / ml IL-13 respectively under the stimulation of 1ng / ml IL-13 were (1222.50 ± 49.95) pg / ml, 1502.83 ± 5.31 pg / ml, 1329.17 ± 56.08 pg / ml and 457.50 ± 15.32 pg / ml respectively, which were significantly higher than that of the control group [(283.50 ± 15.86) pg / ml and , (13.50 ± 5.01) pg / ml, P <0.05 or 0.01] in a concentration-dependent manner. The level of IL-8 protein expression was not significantly different from the control group (P> 0.05). Conclusion IL-13 can stimulate the production of IL-6, MCP-1 and Eotaxin by NHDF. The high expression of these inflammatory factors may be involved in the pathological role of IL-13 in the process of skin fibrosis.
