目的:建立HPLC法测定畲药老虎吊中齐墩果酸的含量。方法:色谱柱为Waters XBridge C18(250 mm×4.6 mm,5μm);流动相为甲醇-0.1 mol·L-1乙酸铵溶液(83∶17);流速为1.0 ml·min-1;柱温为20℃;检测波长为210 nm;进样量为10μl。结果:老虎吊中齐墩果酸与其他组分分离良好,线性范围为72.52~725.2μg·ml-1(r=1.000 0);平均加样回收率为98.05%(RSD=1.89%,n=6)。结论:该方法简便易行、准确、重复性好,可用于畲药老虎吊中齐墩果酸的含量测定。
OBJECTIVE: To establish an HPLC method for the determination of oleanolic acid in Peihuan tiger crane. METHODS: The column was Waters XBridge C18 (250 mm × 4.6 mm, 5 μm). The mobile phase was methanol-0.1 mol·L-1 ammonium acetate solution (83:17), the flow rate was 1.0 ml · min-1. 20 ℃; detection wavelength of 210 nm; injection volume of 10μl. Results: Oleanolic acid was separated from other components in the tiger’s hanging cage with a linear range of 72.52-725.2 μg · ml-1 (r = 1.000 0). The average recovery was 98.05% (RSD = 1.89%, n = 6). Conclusion: The method is simple, accurate, reproducible and can be used for the determination of oleanolic acid in Peihuan tiger crane.
