目的:探讨金黄扶正茶对免疫抑制小鼠脾脏T、B淋巴细胞增殖及腹腔巨噬细胞免疫功能的影响。方法:金黄扶正茶高、中、低剂量组分别连续给昆明种小鼠ig 10 d,同时隔日sc注射环磷酰胺(30 mg·kg-1)造成免疫抑制模型。采用四甲基偶氮唑蓝(MTT)法测定刀豆蛋白A(Con A)诱导脾脏T淋巴细胞增殖和脂多糖(LPS)诱导B淋巴细胞增殖的影响及自然杀伤细胞(NK细胞)和淋巴因子激活的杀伤细胞(LAK细胞)活性,酶法检测腹腔巨噬细胞分泌一氧化氮合酶(NOS)的活性,用全自动酶标仪检测巨噬细胞吞噬中性红能力。结果:金黄扶正茶低、中、高剂量组均能显著提高免疫抑制小鼠的T,B淋巴细胞的吸光度(A,P<0.01);而中、高剂量组能显著提高免疫抑制小鼠NK细胞、LAK细胞活性、腹腔巨噬细胞吞噬活性及诱导型一氧化氮合成酶(iNOS)活力(P<0.01或P<0.05)。结论:金黄扶正茶能促进免疫抑制小鼠脾淋巴细胞增殖,增强NK细胞、LAK细胞活性,提高免疫抑制小鼠腹腔巨噬细胞吞噬活性,从而增强免疫抑制小鼠的细胞免疫功能。
Objective: To investigate the effects of golden huzheng tea on T, B lymphocyte proliferation and immune function of peritoneal macrophages in immunosuppressed mice. Methods: Kunming Fuzheng high, medium and low dose groups were administered to Kunming mice ig 10 d, while every other day sc injection of cyclophosphamide (30 mg · kg-1) caused by immunosuppression model. The effects of Con A on the proliferation of splenic T lymphocytes and the proliferation of B lymphocytes induced by lipopolysaccharide (LPS) were measured by MTT assay and the effects of natural killer (NK) cells and lymph Factor-activated killer cells (LAK cells). The activity of nitric oxide synthase (NOS) secreted by peritoneal macrophages was detected by enzymatic method. The phagocytosis of neutral phagocytes by macrophages was detected by using an automatic microplate reader. Results: The golden, the middle and the high dose of Fuzheng Tea significantly increased the absorbance of T and B lymphocytes in immunosuppressed mice (A, P <0.01) LAK cell activity, phagocytic activity of peritoneal macrophages and iNOS activity (P <0.01 or P <0.05). CONCLUSION: Golden Fuzheng Tea can promote the proliferation of immunosuppressed mice splenic lymphocytes, enhance the activity of NK cells and LAK cells, and enhance the phagocytic activity of peritoneal macrophages in immunosuppressed mice, and enhance the cellular immune function of immunosuppressed mice.
