目的研究2型糖尿病患者Foxa2基因表达与胰岛β细胞功能的关系。方法选取2型糖尿病患者70例为T2DM组,健康体检者65例为对照组,两组患者入选后对一般资料及临床生化指标进行测量和检测,并对T2DM组患者进行Foxa2基因检测,把所测得序列与GenBank提供的外显子和启动子进行比较与分析以找出是否有突变。结果 T2DM组胰岛素抵抗指数高于对照组,而胰岛素敏感指数、胰岛β细胞功能指数、糖负荷后胰岛素增值与血糖增的比值(△I30/△G30)、新β细胞功能指数则低于对照组,差异有统计学意义(P<0.05)。T2DM组经Foxa2基因检测见突变者总计55例(78.6%),Foxa2基因存在突变者胰岛素抵抗指数高于突变阴性者,而胰岛素敏感指数、胰岛素抵抗指数、新β细胞功能指数低于对照组,差异有统计学意义(P<0.05)。结论 2型糖尿病患者Foxa2基因突变可能会增加胰岛素抵抗情况,影响胰岛β细胞功能,由此提示Foxa2基因突变可能为2型糖尿病发病的危险因素之一。
Objective To study the relationship between Foxa2 gene expression and pancreatic β-cell function in type 2 diabetic patients. Methods Totally 70 T2DM patients with type 2 diabetes mellitus and 65 healthy controls were selected as control group. The general data and clinical biochemical parameters were measured and detected in both groups. Foxa2 gene was detected in T2DM patients, The measured sequences are compared with the exons and promoters provided by GenBank to find out if there are any mutations. Results The insulin resistance index of T2DM group was higher than that of the control group, but the ratio of insulin sensitivity index, islet β cell function index, the ratio of insulin value to blood glucose increase (△ I30 / △ G30) and new β cell function index , The difference was statistically significant (P <0.05). In T2DM group, a total of 55 mutations (78.6%) were detected by Foxa2 gene mutation. The insulin resistance index of mutant Foxa2 gene was higher than that of mutant negative gene. The insulin sensitivity index, insulin resistance index and new β cell function index were lower than those of control group The difference was statistically significant (P <0.05). Conclusions Mutations of Foxa2 gene in type 2 diabetic patients may increase insulin resistance and affect the function of pancreatic β-cell. Therefore, Foxa2 gene mutation may be one of the risk factors for type 2 diabetes.
