目的从生物信息学角度分析刚地弓形虫磷酸甘油酸酯变位酶2(TgPGAM2)基因编码蛋白的结构与抗原表位,预测该蛋白的免疫原性。方法利用蛋白分析专家系统(ExPASy)提供的ProtParam、SOSUI、TMHMM、HNN、MotifScan,NCBI上的ORF finder、SignalP、Bcepred等生物信息学在线分析程序,结合Gene Runner、DNAMAN等生物信息学软件,分析、预测TgPGAM2蛋白的理化性质、可溶性、表面可及性?可塑性,信号肽,跨膜区,翻译后修饰位点、亲(疏)水性、二级结构、抗原表位等。结果该蛋白由264个氨基酸组成,分子式为C1345H2097N363O385S11,分子质量为29.865 3 ku,等电点理论值为6.46,波长280 nm时的吸光度(A)值为1.697,半衰期为30 h,有10个表面可及性参数≥1.9的区域、3个亲水性参数得分≥1.9的区域、3个柔韧性参数得分≥2的区域、8个翻译后修饰位点、16个潜在抗原表位,为可溶性表达蛋白。结论刚地弓形虫PGAM2基因编码的Tg PGAM2为可溶性蛋白,具有免疫原性,可作为弓形虫病疫苗候选抗原。
OBJECTIVE: To analyze the structure and epitopes of the encoded protein of T.gondii gondii TgPGAM2 from the perspective of bioinformatics and to predict its immunogenicity. Methods Bioinformatics online analysis programs such as ProtParam, SOSUI, TMHMM, HNN, MotifScan, NCBI, SignalP and Bcepred provided by ExPASy and bioinformatics software such as Gene Runner and DNAMAN Predicting the physicochemical properties, solubility, surface accessibility of TgPGAM2 protein, plasticity, signal peptide, transmembrane region, posttranslational modification sites, pro (hydrophobic), secondary structure, epitopes and so on. Results The protein was composed of 264 amino acids with the molecular formula of C1345H2097N363O385S11, the molecular mass of 29.865 3 ku and the theoretical value of the isoelectric point of 6.46. The absorbance (A) of the protein at 280 nm was 1.697 and the half-life was 30 h. Three regions with score of ≥1.9 in hydrophilic parameter, three regions with score of ≥2 in flexibility parameter, eight post-translational modification sites and sixteen potential epitopes were soluble expression protein. Conclusion Tg PGAM2 encoded by PGAM2 gene of Toxoplasma gondii is a soluble protein that is immunogenic and can be used as a candidate antigen for toxoplasmosis vaccine.
