目的:探讨Notch1、Notch3及Hes1在胃肠道间质瘤(GIST)中的表达及临床意义。方法:收集48例手术切除的原发性GIST组织标本以及48例GIST瘤旁胃肠组织标本,用免疫组化法检测Notch1、Notch3及下游靶基因Hes1蛋白的表达,并分析各蛋白表达与GIST患者临床病理因素的关系。结果:与瘤旁胃肠组织比较,GIST组织中Notch1(58.33%vs.29.17%)、Notch3(97.97%vs.35.47%)、Hes1(35.47%vs.10.42%)蛋白的阳性表达率均明显升高(均P<0.05)。将GIST患者按性别、年龄、肿瘤部位、肿瘤大小及NIH危险分级分组比较各蛋白的表达阳性率,结果显示,Notch1蛋白表达与GIST的NIH危险分级有关(χ2=16.53,P=0.001);Notch3蛋白的表达与以上各因素级均无关(均P>0.05);Hes1蛋白表达与GIST的大小有关(χ2=7.78,P=0.02)。结论:Notch信号通路相关蛋白在GIST组织中表达升高,且可能在GIST的发生、发展中起重要作用。
Objective: To investigate the expression and clinical significance of Notch1, Notch3 and Hes1 in gastrointestinal stromal tumors (GIST). Methods: 48 cases of primary GIST tissue specimens and 48 cases of GIST tissues were collected. Immunohistochemistry was used to detect the expression of Notch1, Notch3 and downstream target gene Hes1. The expression of GIST The relationship between patients’ clinical and pathological factors. Results: The positive expression rates of Notch1 (58.33% vs.29.17%), Notch3 (97.97% vs.35.47%) and Hes1 (35.47% vs.10.42%) in GIST tissues were significantly higher than those in para-tumorous gastrointestinal tissues High (all P <0.05). The GIST patients were grouped by sex, age, tumor location, tumor size and NIH risk grade. The results showed that the expression of Notch1 protein was related to the NIH risk classification of GIST (χ2 = 16.53, P = 0.001) The expression of Hes1 protein was related to the size of GIST (χ2 = 7.78, P = 0.02). CONCLUSION: The expression of Notch signaling pathway related proteins in GIST tissues may play an important role in the development of GIST.
