目的:研究B细胞恶性肿瘤[多发性骨髓瘤(MM)和B细胞非霍奇金淋巴瘤(B-NHL)]患者外周血单个核细胞(PBMC)Blys表达情况,探讨Blys的转录水平与MM、B-NHL的关系,分析Blys在MM和B-NHL发病中的作用。方法:应用实时荧光定量PCR法(RFQ-PCR)检测MM(24例)、B-NHL(104例)患者PBMCBlys mRNA的表达水平,以健康献血员(30例)作为对照,并对B-NHL不同分型及化疗前、后患者PBMCBlys mRNA进行比较。结果:MM、Ⅰ、Ⅱ期和Ⅲ、Ⅳ期B-NHL化疗前PBMCBlys mRNA均显著高于正常对照组,P<0.01;Ⅰ、Ⅱ期和Ⅲ、Ⅳ期B-NHL化疗前均显著高于化疗后,P<0.01,Ⅲ、Ⅳ期B-NHL化疗前显著高于Ⅰ、Ⅱ期,P<0.05。且B-NHL的不同组织类型化疗前PBMCBlysmRNA各不相同,均显著高于正常对照组,P<0.01,且显著高于化疗后,P<0.01;其中,弥漫性大B细胞淋巴瘤显著高于B-慢性淋巴细胞白血病/小细胞淋巴瘤、套细胞淋巴瘤和边缘区淋巴瘤,P值均<0.01。结论:PBMCBlysmRNA的表达水平在MM和B-NHL患者外周血中明显升高,且在B-NHL中与组织类型、分期和治疗有关,提示Blys可能参与B细胞恶性肿瘤发病过程,有望成为B细胞恶性肿瘤诊断、判断分期和评价疗效的新指标。
OBJECTIVE: To study the expression of Blys in peripheral blood mononuclear cells (PBMCs) from patients with B cell malignancies (MM and B-NHL) and to explore the relationship between Blys transcription and MM , B-NHL, analyze the role of Blys in the pathogenesis of MM and B-NHL. Methods: The expression of PBMCBlys mRNA in MM (24 cases) and B-NHL (104 cases) was detected by real-time quantitative PCR (RFQ-PCR). Healthy blood donors (30 cases) PBMCBlys mRNA was compared between different types and before and after chemotherapy. Results: The expression of Blys mRNA in MM, stage Ⅰ, Ⅱ and stage Ⅲ, Ⅳ B-NHL before chemotherapy were significantly higher than those in normal control group, P <0.01. The levels of Bly NHL in stage Ⅰ, Ⅱ and Ⅲ, Ⅳ before chemotherapy were significantly higher than those before B-NHL After chemotherapy, P <0.01, Ⅲ, Ⅳ B-NHL before chemotherapy was significantly higher than Ⅰ, Ⅱ, P <0.05. The expression of BlysmRNA in different tissue types of B-NHL before chemotherapy was significantly different from that in normal control group (P <0.01), and significantly higher than that in chemotherapy group (P <0.01). Among them, diffuse large B-cell lymphoma was significantly B-Chronic lymphocytic leukemia / small cell lymphoma, mantle cell lymphoma and marginal zone lymphoma, both P <0.01. Conclusion: The expression of PBMCBlysmRNA in peripheral blood of patients with MM and B-NHL is significantly higher than that of normal tissues, and it is related to the type, stage and treatment of B-NHL, suggesting that Blys may be involved in the pathogenesis of B-cell malignancies and is expected to become B cells Malignant tumor diagnosis, judgment staging and evaluation of new indicators of efficacy.
