目的:探讨祁州漏芦水提物(RUWE)对小鼠H22移植瘤的生长抑制作用。方法:建立小鼠皮下H22移植瘤模型,将实验动物分为模型组、RUWE高、中、低剂量组和5-Fu组,RUWE按400,200,100 mg.kg-1.d-1剂量ig连续给药10 d,5-Fu组按25 mg.kg-1.d-1剂量隔日ip给药,处死动物,计算抑瘤率、胸腺指数和脾指数,并以ELISA法检测血清肿瘤坏死因子-α(TNF-α)和白介素-2(IL-2),比色法检测血清总抗氧化活力(T-AOC)和丙二醛(MDA)含量。结果:与模型组相比,RUWE能显著减小移植瘤瘤重,提高抑瘤率,其RUWE高、中、低剂量组抑癌率分别为38.05%,34.98%,26.95%。同时,明显升高荷瘤小鼠的脾指数,增高血清IL-2和降低TNF-α,增高血清T-AOC和降低MDA水平。结论:RUWE对H22移植瘤具有明显的抑制作用,其作用可能与其增高机体免疫机能和抗氧化能力有关。
Objective: To investigate the inhibitory effect of water extract from Qizhou (RUWE) on the growth of mouse H22 xenografts. Methods: The subcutaneous H22 xenograft model of mice was established. The experimental animals were divided into model group, RUWE high, medium and low dose groups and 5-Fu group. RUWE was administered continuously at ig 400,200,100 mg.kg-1.d-1 On the 10th day, the animals in the 5-Fu group were administered with 25 mg.kg-1.d-1 every other day. The animals were sacrificed and the tumor inhibition rate, thymus index and spleen index were calculated. The levels of serum tumor necrosis factor- TNF-α and IL-2, and the levels of T-AOC and MDA were measured by colorimetric method. Results: Compared with the model group, RUWE could significantly reduce the tumor weight and increase the tumor inhibition rate. The tumor suppression rates of RUWE high, medium and low dose groups were 38.05%, 34.98% and 26.95%, respectively. At the same time, the spleen index, the serum IL-2 and the TNF-α, the serum T-AOC and the MDA in the tumor-bearing mice were significantly increased. Conclusion: RUWE has a significant inhibitory effect on H22 xenografts, which may be related to the enhancement of immune function and antioxidant capacity.
