目的研究NF-κB在宫颈癌DEK基因沉默中作用。方法实验分转染psiRNA-hHDEK组、转染psiRNA-hH1neo组及未转染组,采用免疫细胞化学染色和western blot方法,观察三组细胞NF-κB的表达情况。结果 psiRNA-hHDEK转染组CaSki细胞NF-κB蛋白表达明显增加;经显微图像分析测定转染psiRNA-hHDEK组阳性细胞百分率明显高于未转染组[(74.2±8.1)%vs(11.7±2.6)%]和转染psiRNA-hH1neo组[(74.2±8.1)%vs(21.6±3.9)%]。结论 DEK基因沉默后CaSki细胞内NF-κB表达上调,DEK负性调控宫颈癌细胞中NF-κB表达,从而发挥其衰老抑制基因及凋亡抑制基因的作用。
Objective To study the role of NF-κB in the gene silencing of cervical cancer. Methods The psiRNA-hHDEK group was transfected into the psiRNA-hH1neo group and the untransfected group. Immunocytochemistry and western blot were used to observe the expression of NF-κB in the three groups of cells. Results The expression of NF-κB protein in psiRNA-hHDEK transfected CaSki cells was significantly increased. The percentages of positive cells transfected with psiRNA-hHDEK were significantly higher than those in untransfected cells [(74.2 ± 8.1)% vs (11.7 ± 2.6%] and psiRNA-hH1neo transfection [(74.2 ± 8.1)% vs (21.6 ± 3.9)%]. Conclusions DEK gene silencing can up-regulate the expression of NF-κB in CaSki cells. DEK negatively regulates the expression of NF-κB in cervical cancer cells, thereby exerting the effects of aging suppressor gene and apoptosis-suppressing gene.
