目的研究单纯疱疹病毒2 型(HSV-2)对人单核细胞表达HLA-II 类抗原(HLA-DR、HLA-DQ)的影响,探讨单核细胞及HLA-II类抗原在单纯疱疹病毒2型感染中的作用。方法用单纯疱疹病毒2型(333株)感染人单核细胞,于感染后1、3、5、7 天分别收集细胞,用APAAP 法检测单核细胞HLA-DR、HLA-DQ表达水平。结果单纯疱疹病毒2 型感染后1 天,单核细胞表达HLA-KR、HLA-DQ 抗原水平明显降低,以后逐渐升高,至7 天达到对照组水平。结论单纯疱疹病毒2 型感染早期能抑制单核细胞表达HLA-II类抗原,HSV-2抑制单核细胞表达HLA-II类抗原可能是HSV-2入侵机体的手段之一。HLA-II类抗原在单纯疱疹病毒2型致病及免疫过程中可能起重要作用。
Objective To investigate the effect of herpes simplex virus type 2 (HSV-2) on the expression of HLA class II antigens (HLA-DR, HLA-DQ) in human monocytes and to explore the role of monocytes and HLA class II antigens in herpes simplex virus 2 Type infection in the role. Methods Human herpes simplex virus type 2 (333) was used to infect human monocytes. Cells were harvested at 1, 3, 5 and 7 days after infection. The expression of HLA-DR and HLA-DQ was detected by APAAP. RESULTS: On the 1st day after HSV-2 infection, HLA-KR was expressed in monocytes. The level of HLA-DQ antigen was significantly decreased, and then gradually increased until reaching the level of control after 7 days. Conclusions HSV-2 can inhibit the expression of HLA-class II antigens in monocytes during the early stages of HSV-2 infection, and HSV-2 inhibits the expression of HLA-II antigens in monocytes. HLA class II antigens may play an important role in the herpes simplex virus type 2 pathogenicity and immune processes.
