目的探讨镉恶性转化人支气管上皮细胞(16HBE)中各种翻译启动因子的表达变化。方法利用非转化16HBE对照细胞和氯化镉(CdCl2)恶性转化16HBE成瘤细胞,通过逆转录-聚合酶链反应(RT-PCR)及荧光定量PCR(FQ-PCR)等方法,观察与分析CdCl2恶性转化16HBE细胞中翻译启动因子家族mRNA的表达变化情况。结果以β-actin作为内参照,相对于对照细胞,在CdCl2转化16HBE成瘤细胞23个翻译启动因子的特异片段中,RT-PCR检测结果显示eIF2a、eIF2C1、eIF4E因子的mRNA表达水平均有不同程度的升高,而eIF2β、eIF4EBP因子的mRNA表达量则有不同程度的下降;FQ-PCR结果证实,镉恶性转化成瘤细胞中,eIF2a、eIF2C1、eIF4E的表达水平相对于对照细胞分别升高22倍、21倍和325倍,而eIF2β、eIF4EBP因子的表达量分别是对照细胞的0.005倍和0.107倍。结论研究结果提示,镉的致癌作用可能涉及到多个翻译因子的异常表达。
Objective To investigate the expression changes of various translational promoters in cadmium malignant transformed bronchial epithelial cells (16HBE). Methods Non-transformed 16HBE control cells and cadmium chloride (CdCl2) malignant transformation of 16HBE into tumor cells, by reverse transcription - polymerase chain reaction (RT-PCR) and quantitative PCR (FQ-PCR) and other methods to observe and analyze CdCl2 Expression of translation initiation factor family mRNA in malignant transformed 16HBE cells. Results β-actin was used as an internal reference, and RT-PCR results showed that the expression levels of eIF2a, eIF2C1 and eIF4E mRNA were different in the specific fragments of 23 translation promoters of 16HBE tumor cells transformed by CdCl2 compared with control cells While the expression of eIF2β and eIF4EBP decreased in some degree. The results of FQ-PCR confirmed that the expression levels of eIF2a, eIF2C1 and eIF4E in malignant transformed cadmium were higher than those in control cells 22-fold, 21-fold and 325-fold, respectively. The expression levels of eIF2β and eIF4EBP were 0.005 times and 0.107 times that of the control cells, respectively. Conclusion The results suggest that the carcinogenicity of cadmium may involve the abnormal expression of multiple translation factors.
