目的探讨非小细胞肺癌(non-small lung cancer,NSCLC)中survivin和bcl-xl基因蛋白的表达及其相关性。方法选择2004年2月至2005年10月聊城市人民医院收治的32例NSCLC患者,应用组织芯片技术制成32例NSCLC组织芯片,采用免疫组织化学技术检测同期30名正常成人肺组织及32例NSCLC患者肺组织中survivin及bcl-xl基因蛋白的表达。结果survivin和bcl-xl基因蛋白在NSCLC组织中表达阳性率分别为71·9%和81·3%,显著高于正常肺组织(P<0·05)。survivin蛋白表达与肺癌TNM分期、淋巴结转移及细胞分化程度密切相关,但与肿瘤组织学类型、年龄、性别无关(P>0·05)。bcl-xl基因蛋白表达与细胞分化程度密切相关(P<0·05),与肿瘤TNM分期、淋巴结转移、肿瘤组织学类型、年龄、性别无关(P>0·05)。survivin与bcl-xl基因蛋白过表达呈正相关(P<0·05)。结论survivin和bcl-xl基因蛋白在NSCLC中表达增强且呈正相关,可能与NSCLC发生有关,survivin在NSCLC淋巴结转移方面可能起着重要作用,提示NSCLC预后不良。
Objective To investigate the expression of survivin and bcl-xl protein in non-small lung cancer (NSCLC) and their correlation. Methods Thirty-two patients with NSCLC admitted from Liaocheng People’s Hospital from February 2004 to October 2005 were selected and 32 cases of NSCLC tissue microarray were obtained by tissue microarray. 30 normal adult lung tissues and 32 cases of normal lung tissue were detected by immunohistochemistry. Expression of survivin and bcl-xl in lung tissue of NSCLC patients. Results The positive rates of survivin and bcl-xl protein in NSCLC tissues were 71.9% and 81.3%, respectively, which were significantly higher than those in normal lung tissues (P <0.05). The expression of survivin protein was closely related to the TNM stage, lymph node metastasis and cell differentiation in lung cancer, but not to histological type, age and sex (P> 0.05). The expression of bcl-xl protein was closely related to the degree of cell differentiation (P <0.05), which was not associated with tumor TNM stage, lymph node metastasis, tumor histological type, age and sex (P> 0.05). Survivin and bcl-xl gene protein overexpression was positively correlated (P <0.05). Conclusion The expressions of survivin and bcl-xl protein are positively correlated with the expression of bcl-xl protein in NSCLC, which may be related to the occurrence of NSCLC. Survivin may play an important role in lymph node metastasis of NSCLC, suggesting poor prognosis of NSCLC.
