目的观察T辅助细胞17(Th17)相关细胞因子白细胞介素17(IL-17)和白细胞介素23(IL-23)在实验性自身免疫性神经炎(EAN)模型中的表达,探讨Th17细胞在EAN的作用。方法用牛周围神经鞘磷脂(BPM)免疫Lewis大鼠,观察免疫后9、15、21d大鼠的临床表现和坐骨神经中IL-17、IL-23蛋白及mRNA的表达。结果免疫后9dEAN大鼠开始出现临床症状,15d到达疾病高峰。IL-17蛋白及mRNA在免疫后第15天表达均达到最高点,以后逐渐下降,且各组与对照组相比差异显著(P<0.05)。IL-23蛋白及mRNA在免疫后9d表达最多,随后逐渐下降,9、15d与对照组相比差异显著(P<0.05)。结论IL-17可能与EAN的疾病进程有关,IL-23可能在EAN疾病诱导阶段起一定作用。
Objective To observe the expression of Th17 related cytokines, interleukin 17 (IL-17) and interleukin 23 (IL-23), in experimental autoimmune neuritis (EAN) The role of EAN. Methods Lewis rats were immunized with bovine peripheral sphingomyelin (BPM) to observe the clinical manifestations and the expression of IL-17, IL-23 protein and mRNA in the sciatic nerve of rats at 9, 15 and 21 days after immunization. Results After 9 days of immunization, rats began to show clinical symptoms and peaked at 15 days. The expression of IL-17 protein and mRNA reached the highest point on the 15th day after immunization, and then decreased gradually. The difference between each group and the control group was significant (P <0.05). The highest expression of IL-23 protein and mRNA was detected 9 d after immunization, and then decreased gradually. The difference was significant at 9 and 15 d (P <0.05). Conclusion IL-17 may be related to the disease process of EAN, and IL-23 may play a role in the induction stage of EAN.
