目的 探讨中药复方益气解毒颗粒对鼻咽癌细胞杀伤作用的分子机制。方法 筛选益气解毒颗粒药液对鼻咽癌杀伤的半抑制浓度(IC_(50)),用0.1倍半抑制浓度(0.1×IC_(50))的益气解毒颗粒作用鼻咽癌细胞HNE1 96h,用固相pH梯度双向凝胶电泳分离益气解毒颗粒处理和未处理的鼻咽癌细胞HNE1的总蛋白质,银染显色,PDQuest 2-de软件分析差异蛋白质点。结果 益气解毒颗粒药液杀伤鼻咽癌细胞HNE1的半抑制浓度为6.25mg/ml;双向电泳图像分析益气解毒颗粒药物处理组图像的平均蛋白质数为1120±89,未处理组的平均蛋白质数为1281±102;差异蛋白质有673个,其中有218个表达增强,有455个蛋白质表达下降。结论 益气解毒颗粒对鼻咽癌细胞的杀伤作用这些差异表达蛋白质有关,这些差异蛋白质有待质谱分析鉴定,为中药作用的分子机理研究提供了新的思路。
Objective To investigate the molecular mechanism of Chinese herb compound Yiqi Jiedu Granule on nasopharyngeal carcinoma cells. Methods The half-inhibitory concentration of Yiqi Jiedu Granule on nasopharyngeal carcinoma cells was screened (IC 50), and treated with Yiqi Jiedu Granules (0.1 × IC 50) The total protein of HNE1 cells treated with Yiqi Jiedu Granules and untreated nasopharyngeal carcinoma cells were separated by two-dimensional gel electrophoresis with solid-phase pH gradient. Silver staining was performed on the cells. PDQuest 2-de software was used to analyze the differential protein spots. Results The half-inhibitory concentration of Yiqi Jiedu Granules on the HNE1 cells was 6.25mg / ml. The average number of proteins in Yiqi Jiedu Granules group treated with two-dimensional electrophoresis was 1120 ± 89. The mean The number of proteins was 1281 ± 102; the number of differential proteins was 673, of which 218 were increased and 455 were decreased. Conclusions The killing effect of Yiqi Jiedu Granules on nasopharyngeal carcinoma cells is related to these differentially expressed proteins. These differential proteins need to be identified by mass spectrometry, providing new ideas for the molecular mechanism of traditional Chinese medicine.
