本研究在人类鼻咽癌(NPC)活检组织及NPC转移淋巴结中发现一种新的DNA多聚酶,它有如下特点:(1)可被KCl及(NH_4)_2SO_4所激活。(2)此酶存在于NPC胞核中。(3)在低分化及未分化型NPC胞核此酶能较好地利用合成模板引物Poly(dA)·oligo(dT)_(12-18)。这种酶与正常人胚鼻咽上皮细胞及Raji细胞中DNA多聚酶的性质不同,而与Burkitt淋巴瘤的EBV-DNA多聚酶性质相似。 本文用NPC活检组织制成的粗酶提取液检测此种EBV-DNA多聚酶,结果在26例中有20例阳性(77%)。文中对NPC组织中含有此EBV-DNA多聚酶的意义进行了讨论。
In this study, a novel DNA polymerase was discovered in the human nasopharyngeal carcinoma (NPC) biopsy and NPC metastatic lymph nodes. It has the following characteristics: (1) It can be activated by KCl and (NH_4)_2SO_4. (2) This enzyme is present in the NPC nucleus. (3) In the poorly differentiated and undifferentiated NPC nucleus, this enzyme can use the synthetic template primer Poly(dA)·oligo(dT)_(12-18). This enzyme has different properties from DNA polymerase in normal human embryonic nasopharyngeal epithelial cells and Raji cells, and is similar in properties to Burkitt’s lymphoma EBV-DNA polymerase. In this paper, the EBV-DNA polymerase was detected using crude enzyme extracts made from NPC biopsies. Results showed that 20 out of 26 cases were positive (77%). The significance of this EBV-DNA polymerase in NPC tissue was discussed.
