目的探讨mRNA差异表达片段与肺血栓栓塞症的关系。方法提取于2001年9月至2002年4月首都医科大学附属北京安贞医院呼吸内科收治的肺血栓栓塞症患者11例和健康对照者11名外周血单个核细胞总RNA,筛选肺动脉血栓栓塞患者差异表达的mRNA片段,并进行表达量鉴定和同源性比较。结果共分离、筛选出55条差异表达条带,选择其中10条(与对照组相比,上调或下调3倍以上)进行克隆测序分析,与GenBank进行读框分析比较,从中得到2个同源性低的未知基因,证实为新基因片段,分别命名为g44、g48;以g44为探针,进行Northern杂交,发现此片段在对照组及疾病组均有杂交信号,但疾病组杂交信号明显强于对照组的杂交信号。结论g44、g88,此片段在肺动脉血栓栓塞疾病组有较强的表达,提示其表达可能与疾病的发生发展密切相关。
Objective To investigate the relationship between mRNA differential expression fragments and pulmonary thromboembolism. Methods Total RNA of peripheral blood mononuclear cells from 11 patients with pulmonary thromboembolism and 11 healthy controls were collected from Department of Respiratory Medicine, Beijing Anzhen Hospital, Capital Medical University from September 2001 to April 2002 and were screened for pulmonary thromboembolism Differentially expressed mRNA fragments were identified and their homology was compared. Results A total of 55 differentially expressed bands were screened and 10 of them were selected (up-regulated or down-regulated more than 3-fold compared with the control group) for cloning and sequencing analysis. Compared with GenBank, two homologous The unknown gene with low sex was confirmed as a new gene fragment, named g44, g48 respectively. Northern blotting with g44 probe showed that this fragment had hybridization signal in the control group and disease group, but the hybridization signal in the disease group was significantly stronger Hybridization signal in control group. Conclusion g44, g88, this fragment in the pulmonary thromboembolism disease group have strong expression, suggesting that the expression may be closely related to the occurrence and development of the disease.
