目的:建立兔髂动脉球囊损伤模型观察复方丹参滴丸(CDDP)对损伤内膜增生修复的影响。方法:健康新西兰雄性大白兔24只随机分为CDDP组和对照组。CDDP组以CDDP(300mg/d)灌喂,对照组给予同等量淀粉片,7d后球囊扩张损伤髂动脉。分别于术前,术后1、2、4周耳缘静脉采血检测一氧化氮(NO)浓度;4周后提取损伤动脉标本,观察内皮修复、内膜增生情况并测定病理形态学参数。结果:CDDP组术后2、4周NO浓度明显高于对照组[(92.12±12.63)∶(82.05±12.24)μmol/L;(97.86±10.14)∶(83.27±13.11)μmol/L];术后4周病理组织学见CDDP组内膜增生程度较对照组明显减轻,新生内膜中血管平滑肌细胞和纤维组织较对照组明显减少,内皮较完整、光滑,管腔狭窄程度较轻。形态学参数见CDDP组的管腔面积明显大于对照组[(1.29±0.31)∶(0.94±0.24)mm2,均P<0·05],新生内膜面积与内膜增生百分比明显小于对照组[(0.85±0.33)∶(1.18±0·36)mm2;(40±7)%∶(55±6)%,均P<0·05]。结论:CDDP能促进损伤内膜修复,减少内膜增生,降低再狭窄率。
OBJECTIVE: To establish a rabbit radial artery balloon injury model and observe the effects of Compound Danshen Dripping Pill (CDDP) on the repair of damaged intimal hyperplasia. Methods: Twenty-four healthy New Zealand white rabbits were randomly divided into CDDP group and control group. The CDDP group was fed with CDDP (300 mg/d), and the control group was given the same amount of starch tablets. After 7 days, the balloon inflated the injured iliac artery. The blood concentration of nitric oxide (NO) was measured before and 1, 2, and 4 weeks after surgery. The injured artery was extracted after 4 weeks to observe the repair of the endothelium and the intimal hyperplasia and to determine the pathological parameters. Results: The NO concentration in the CDDP group was significantly higher than that in the control group [(92.12±12.63):(82.05±12.24)μmol/L; (97.86±10.14):(83.27±13.11)μmol/L]; After 4 weeks of histopathology, the degree of intimal hyperplasia in the CDDP group was significantly less than that in the control group. The vascular smooth muscle cells and fibrous tissue in the neointima were significantly reduced compared to the control group. The endothelium was relatively intact, smooth, and the lumen stenosis was mild. For morphological parameters, the luminal area of the CDDP group was significantly greater than that of the control group [(1.29±0.31):(0.94±0.24) mm2, all P<0.05]. The percentage of neointimal hyperplasia and neointimal hyperplasia was significantly lower than that of the control group [ (0.85±0.33): (1.18±0.36) mm2; (40±7)%: (55±6)%, all P<0.05. Conclusion: CDDP can promote the repair of damaged intima, reduce intimal hyperplasia, and reduce the rate of restenosis.
