目的 观察胆囊收缩素八肽 (CCK -8)对谷氨酸神经毒性作用所致神经元病理损害的影响。 方法 采用扫描及透射电镜技术分别观察经不同因素处理后的原代培养的大鼠大脑皮层神经元和经侧脑室注射药物后的大鼠海马CA1 区神经元的形态学变化 ,同时应用生物体视学计量分析法对细胞内线粒体各指标的变化进行比较。 结果 CCK -8在体内外均能明显改善谷氨酸所致神经元胞体、细胞膜肿胀及破溃 ,轴、树突断裂、丢失或线粒体肿胀、数目减少等病理损害。 结论 CCK -8在体内外具有拮抗谷氨酸神经毒性作用
Objective To observe the effect of cholecystokinin octapeptide (CCK-8) on neuronal pathological damage induced by glutamate neurotoxicity. Methods The morphological changes of hippocampal CA1 neurons in primary cultured rat cortical neurons treated by different factors and by intracerebroventricular injection of drugs were observed by scanning and transmission electron microscopy respectively. At the same time, Econometrics analysis of mitochondrial intracellular each index changes. Results Both CCK-8 and CCK-8 could significantly ameliorate the pathological damage of glutamate-induced neuronal somatic cell bodies, cell membrane swelling and rupture, axon and dendritic rupture, loss or mitochondrial swelling and the number decreased. Conclusion CCK -8 antagonizes glutamate neurotoxicity both in vitro and in vivo
