目的研究DNA拓扑异构酶-Ⅱα(Topoisomerase-Ⅱα,Topo-Ⅱα)基因在人脑胶质瘤中的表达改变及其对胶质瘤U87MG细胞凋亡的影响。方法 Real-time PCR检测25例脑胶质瘤及相应癌旁组织中Topo-ⅡαmRNA的表达。将Topo-Ⅱα基因特异性的siRNA转染U87MG细胞。转染后,Western Blot检测转染后Topo-Ⅱα蛋白的表达,双标流式细胞法检测细胞凋亡率。结果与癌旁组织相比,Topo-ⅡαmRNA在脑胶质瘤组织中表达明显上调。转染后,U87MG细胞中的Topo-Ⅱα蛋白表达明显降低,同时,凋亡率明显增加。结论 Topo-Ⅱα基因在脑胶质瘤中存在表达异常,其表达降低能够促进细胞凋亡。
Objective To investigate the changes of Topoisomerase-Ⅱα (Topo-Ⅱα) gene in human glioma and its effect on the apoptosis of glioma U87MG cells. Methods Real-time PCR was used to detect the expression of Topo-Ⅱα mRNA in 25 gliomas and corresponding paracancerous tissues. Topo-IIα gene-specific siRNA was transfected into U87MG cells. After transfection, the expression of Topo-Ⅱα protein was detected by Western Blot and the apoptosis rate was detected by double-labeled flow cytometry. Results The expression of Topo-Ⅱα mRNA was significantly up-regulated in glioma tissues compared with adjacent tissues. After transfection, the expression of Topo-Ⅱα protein in U87MG cells was significantly decreased, meanwhile, the apoptosis rate was significantly increased. Conclusion Topo-Ⅱα gene is abnormally expressed in gliomas, and the decrease of its expression can promote apoptosis.
