目的:研究鲫鱼卵唾液酸糖肽(Ca-SGPP)对成骨细胞MC3T3-E1骨形成活性及OPG/RANKL基因表达水平的影响。方法:在MC3T3-E1细胞中加入不同浓度的Ca-SGPP,采用MTT法、试剂盒和茜素红染色法检测Ca-SGPP对MC3T3-E1细胞增殖、分化和矿化的影响;利用逆转录聚合酶链式反应(RT-PCR)技术检测MC3T3-E1细胞骨形态发生蛋白-2(BMP-2)、骨保护素(OPG)及核因子κB受体活化因子配体(RANKL)基因m RNA表达水平。结果:Ca-SGPP可显著促进MC3T3-E1细胞增殖及ALP、OCN和COL I的分泌,提高MC3T3-E1细胞矿化能力,上调其BMP-2表达水平及OPG/RANKL的比值,且分子质量大的糖肽效果更佳。结论 :CaSGPP具有促进MC3T3-E1细胞增殖、分化和矿化的能力,其机制可能与OPG/RANKL比值上调有关。
OBJECTIVE: To study the effect of Ca-SGPP on bone formation activity and OPG / RANKL gene expression of osteoblast MC3T3-E1. Methods: Different concentrations of Ca-SGPP were added to MC3T3-E1 cells. The effects of Ca-SGPP on the proliferation, differentiation and mineralization of MC3T3-E1 cells were detected by MTT assay, kit and alizarin red staining. The mRNA expression of bone morphogenetic protein-2 (BMP-2), osteoprotegerin (OPG) and RANKL gene in MC3T3-E1 cells was detected by RT-PCR Level. Results: Ca-SGPP could significantly promote the proliferation of MC3T3-E1 cells and the secretion of ALP, OCN and COLI, increase the mineralization ability of MC3T3-E1 cells, up-regulate the expression of BMP-2 and the ratio of OPG / RANKL, Glycopeptide effect is better. CONCLUSIONS: CaSGPP can promote the proliferation, differentiation and mineralization of MC3T3-E1 cells. The mechanism may be related to the up-regulation of OPG / RANKL ratio.
