目的探讨血管合成血管紧张素Ⅱ及血管合成醛固酮在高血压大鼠中的变化。方法用肠系膜动脉离体灌注,在灌注液中测定血管紧张素Ⅱ及醛固酮。用RT-PCR及Southernblot法检测醛固酮合成酶基因表达的情况。结果SHR血管合成的血管紧张素Ⅱ及醛固酮增多、主动脉组织内醛固酮合成酶基因mRNA表达增强。结论SHR血管合成的血管紧张素Ⅱ及醛固酮增多,血醛固酮合成酶基因表达上调。
Objective To investigate the changes of vascular synthesis of angiotensin Ⅱ and vascular aldosterone in hypertensive rats. Methods In vitro perfusion of mesenteric artery was used to measure angiotensin Ⅱ and aldosterone in perfusate. The expression of aldosterone synthase gene was detected by RT-PCR and Southern blot. Results Angiotensin II and aldosterone were increased in SHR vessels, and the expression of aldosterone synthase gene in aorta tissues was increased. Conclusion Angiotensin Ⅱ and aldosterone are increased in SHR vessels, and the expression of serum ALD gene is up-regulated.
