目的:探讨姜黄素纳米混悬剂对D-半乳糖所致亚急性衰老模型小鼠的衰老症状改善效果及抗衰老相关指标的影响。方法:将衰老模型小鼠分为6组,空白对照组灌胃给予蒸馏水(0.2 m L/10 g·d),其余各组连续皮下注射D-半乳糖(50 mg/kg·d),连续6 w。从注射第2周开始,高、中、低3个给药组分别给予600 mg/kg、300 mg/kg、100 mg/kg姜黄素纳米混悬剂,阳性对照组给予维生素E(50 mg/kg),模型对照组给予等量的蒸馏水(0.2 m L/10 g),连续灌胃给药5 w,测定各组的学习训练成绩、记忆成绩及血清超氧化物歧化酶(SOD)、乙酰胆碱酯酶(ACHE)、和脑组织丙二醛(MDA)、谷光甘肽过氧化物酶(GSH-Px)、总抗氧化能力(T-AOC)等指标水平变化。结果:姜黄素纳米混悬剂高、中、低3个给药组训练成绩和记忆成绩均明显优于模型组(P<0.05),SOD、T-AOC、ACHE、GSH-Px活力显著优于模型组(P<0.05,P<0.01),MDA含量显著低于模型组(P<0.05)。结论:姜黄素纳米混悬剂能有效改善D-半乳糖所引起的记忆障碍,提高抗氧化能力,防止衰老。
Objective: To investigate the effect of curcumin nanosuspensions on aging symptoms and anti-aging related indexes in sub-acute aging mice induced by D-galactose. Methods: The aging model mice were divided into six groups. The blank control group was given distilled water (0.2 m L / 10 g · d), while the other groups were injected subcutaneously with D-galactose (50 mg / kg · d) 6 w. From the second week after injection, curcumin nanosuspension (600 mg / kg, 300 mg / kg, 100 mg / kg) was given to the three high, middle and low dose groups respectively. The positive control group was given vitamin E (50 mg / kg). The model control group was given equal volume of distilled water (0.2 m L / 10 g) for 5 weeks. The learning and training scores, memory and serum levels of superoxide dismutase (SOD), acetylcholine (ACHE) and malondialdehyde (MDA), glutathione peroxidase (GSH-Px) and total antioxidant capacity (T-AOC) Results: The training scores and memory scores of curcumin nanosuspension group were significantly higher than those of the model group (P <0.05). The activities of SOD, T-AOC, ACHE and GSH-Px were significantly better Model group (P <0.05, P <0.01), MDA content was significantly lower than the model group (P <0.05). CONCLUSION: Curcumin nanosuspensions can effectively improve the memory impairment caused by D-galactose, improve the anti-oxidation ability and prevent aging.
