目的建立小鼠肥大细胞瘤 P815种植瘤模型 ,并对其体内诱发特异性 CTL应答机理进行初步的研究。方法经有限稀释法筛选 P815单克隆瘤系 ,用 RT- PCR及产物 DNA测序鉴定肿瘤抗原 P1A基因的表达 ;在此基础上 ,用活瘤苗免疫同系小鼠 ,经标准 5 1 Cr释放试验检测在体特异性 CTL的活性。结果筛选出了一个 P815单克隆成瘤系 P815 - F3,并鉴定该瘤存在 P1A基因的表达 ;用活瘤疫苗免疫 6只小鼠 ,在 3只体内诱发出了特异性 CTL 应答。结论本研究成功地建立了 P815肿瘤模型 ,对体内特异性抗瘤 CTL效应的诱发和检测做了初步的研究 ,这为以后基于该肿瘤模型的肿瘤疫苗研究及相关肿瘤免疫学基础研究奠定了基础
OBJECTIVE: To establish a mouse model of mast cell tumor P815 tumor and to study its mechanism of inducing specific CTL response in vivo. METHODS: The P815 monoclonal antibody line was screened by limiting dilution method. The expression of tumor antigen P1A gene was identified by RT-PCR and DNA sequencing. On this basis, mice were immunized with live tumor vaccine and tested by standard 51 Cr release assay. In vivo-specific CTL activity. Results One P815 monoclonal tumorigenesis P815-F3 was screened out and the P1A gene expression was identified. Six mice were immunized with live tumor vaccine, and specific CTL responses were induced in 3 of them. Conclusion This study successfully established the P815 tumor model and conducted a preliminary study on the induction and detection of specific anti-tumor CTL effects in vivo. This laid the foundation for the future research on tumor vaccine based on this tumor model and the basic research of related tumor immunology.
