目的:建立大鼠高血脂及脂肪肝模型,研究停止高脂饲料干预后观察其自愈时间。方法:SD大鼠高脂饲料饲养连续3周,测定大鼠血清甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)。然后间隔一周左右测血脂及取部分大鼠肝脏,固定、染色后光镜下进行组织病理学检查。结果:给予高脂饲料3周模型组大鼠血清TG、TC明显高于正常组,高密度脂蛋白胆固醇(HDL-C)/总胆固醇(TC)明显低于正常组。停止高脂饲料干预后第12天、22天、29天,模型组大鼠血清TC明显高于正常组;停止高脂饲料干预后第12天、22天,模型组大鼠血清(HDL-C)/TC明显低于正常组;病理组织学检查表明,模型组大鼠在停止高脂饲料干预后第12天,肝小叶结构模糊不清,部分肝细胞胞浆内出现大小不等的空泡,大部分肝细胞呈气球样变,部分动物肝细胞出现点状或灶状坏死;第22天,可见肝细胞肿胀,轻度脂肪变及气球样变;第29天肝脏组织细胞结构恢复正常。结论:大鼠给予高脂饲料喂养3周,可出现非常明显的高脂血症,停止高脂饲料干预后第22天,模型组动物仍出现高胆固醇血症及脂肪肝,在停止高脂饲料干预4周后各项指标基本恢复正常。
Objective: To establish rat model of hyperlipidemia and fatty liver and to study the time of self-healing after stopping the intervention of high fat diet. On the 22nd day, hepatocytes swell, mild fatty changes and balloon-like changes were observed. On the 29th day, the liver cells returned to normal. Conclusion: Rats fed with high-fat diet for 3 weeks showed very obvious hyperlipidemia. On the 22nd day after stopping high-fat diet, hypercholesterolemia and fatty liver still appeared in the model group, After 4 weeks of intervention, the indexes returned to normal.
