目的:探讨二甲双胍治疗前后多囊卵巢综合征(PCOS)大鼠子宫内膜组织中Hoxa10及胰岛素(INS)、INS受体(INS-R)蛋白表达的变化.方法:选取85日龄SD雌性大鼠54只,随机分为:①对照组(20只);②PCOS组(17只);③二甲双胍治疗组(17只).后两组先参照PoretskyL等方法复制PCOS大鼠模型,造模成功后,PCOS组喂服安慰剂,治疗组喂服二甲双胍,14d后断头处死各组大鼠并取材.采用免疫组织化学ElivisionTMPlus两步法检测三组子宫内膜Hoxa10及INS,INS-R蛋白的表达.并取外周血测定INS和睾酮含量.结果:PCOS组大鼠子宫内膜Hoxa10和INS-R表达下降(P<0.01,P=0.012),INS表达增加(P=0.003);二甲双胍治疗后子宫内膜Hoxa10表达增加(P<0.01),INS水平下降(P=0.043),但均低于对照组水平(P<0.01,P=0.037);INS-R无明显变化;治疗后血INS和血睾酮水平均下降(P<0.01,P=0.048).结论:PCOS大鼠子宫内膜可能存在INS抵抗和容受性下降,治疗后可部分逆转此变化.
Objective: To investigate the changes of Hoxa10, INS and INS-R protein expressions in endometrial tissue of PCOS rats before and after treatment with metformin.Methods: (N = 54) were randomly divided into: ① control group (20); ② PCOS group (17); ③ metformin treatment group (17) .After the two groups first reference PoretskyL and other methods to copy PCOS rat model, , The PCOS group was given placebo and the treatment group was given metformin. After 14 days, the rats in each group were sacrificed and decapitated, and the expression of Hoxa10, INS and INS-R protein in the three groups were detected by immunohistochemical ElivisionTMPlus two-step method .The INS and testosterone contents were measured in peripheral blood.Results: The expressions of Hoxa10 and INS-R in endometrium of PCOS rats decreased (P <0.01, P = 0.012), INS increased (P = 0.003) In endomembrane Hoxa10 expression increased (P <0.01), INS level decreased (P = 0.043), but were lower than the control group (P <0.01, P = 0.037); INS-R no significant change; Testosterone levels decreased (P <0.01, P = 0.048) .Conclusion: There may be decreased INS resistance and tolerance in endometrium of PCOS rats, which may be partially reversed after treatment .
