目的:探讨Ezrin表达水平与肺癌转移潜能的相关性以及细胞内微丝骨架的变化差异。方法:应用蛋白质印迹法和RT-PCR方法检测4种具有不同转移潜能的人类肺癌细胞系中Ezrin mRNA和蛋白的表达水平;荧光染色的方法显示细胞内微丝骨架的变化。结果:Ezrin的mRNA和蛋白表达水平在高转移潜能的人类肺癌细胞中高于相应的低转移肺癌细胞;划痕修复实验显示,高转移组细胞的划痕修复能力明显增强。95D和801D细胞的划痕修复率〔(52.15±5.21)%和(54.22±4.55)%〕明显高于95C和A549细胞的划痕修复率〔(20.15±4.85)%和(21.33±5.01)%〕,差异有统计学意义,P<0.01。高转移性肺癌与低转移肺癌细胞比较,前者细胞内肌动蛋白多聚体减少,微丝骨架杂乱、模糊,而低转移肺癌细胞和肺成纤维细胞微丝骨架粗大、结构清晰、排列整齐。结论:Ezrin mRNA和蛋白表达水平与肺癌的转移能力呈正相关性;肺癌转移潜能的增高伴有肌动蛋白多聚体的丢失和微丝结构的解聚,微丝骨架的这种变化与Ezrin的表达增高具有相关性。
Objective: To investigate the correlation between the expression level of Ezrin and metastasis potential of lung cancer and the difference of intracellular microfilament skeleton. Methods: Western blotting and RT-PCR were used to detect the expression of Ezrin mRNA and protein in four human lung cancer cell lines with different metastatic potentials. Fluorescence staining was used to detect the changes of intracellular microfilament cytoskeleton. Results: The mRNA and protein expression of Ezrin in lung cancer cells with high metastatic potential was higher than that in the corresponding low metastatic lung cancer cells. Scratch repair experiments showed that the scratch repair ability of Ezrin cells was significantly enhanced. Scratch repair rates 〔(52.15 ± 5.21)% and (54.22 ± 4.55)%〕 of 95D and 801D cells were significantly higher than those of 95C and A549 cells 〔(20.15 ± 4.85)% and (21.33 ± 5.01% 〕, The difference was statistically significant, P <0.01. Compared with low-metastatic lung cancer cells, the former reduced the actin multimer in the cells and the microfilament skeleton was disorderly and fuzzy, while the microfilament skeletons in the lung cancer cells and the lung fibroblasts in the low metastatic lung cancer were coarse, with clear structure and neat arrangement. Conclusion: The expression of Ezrin mRNA and protein is positively correlated with the metastasis of lung cancer. The metastatic potential of lung cancer is associated with the loss of actin multimers and depolymerization of microfilament structure. This change of microfilament skeleton is similar to that of Ezrin Increased expression is correlated.
