目的:观察去甲斑蝥素(NCTD)体外对卵巢癌3AO细胞和AO细胞的抑制作用,并初步探讨其作用机制。方法:利用细胞培养技术,以结晶紫染色法测定NCTD对卵巢癌3AO和AO细胞的抑制作用,并利用流式细胞仪测定分析NCTD作用下两种肿瘤细胞的细胞周期变化。结果:NCTD1μg/ml即对3AO和AO细胞有轻微抑制作用,其半数抑制剂量(IC50)分别为20μg/ml和10μg/ml。NCTD作用6h,3AO和AO细胞生长即受到抑制,其抑制率随着药物作用时间延长而升高。在NCTD作用下,3AO和AO细胞G2+M期细胞百分比明显升高,而S期细胞百分比相应地明显降低。结论:NCTD体外对卵巢癌3AO和AO细胞有明显抑制作用,其抑制作用呈剂量和时间依赖性。NCTD抗肿瘤细胞增殖的机制可能与其将肿瘤细胞特异阻抑于G2+M期有关
Objective: To observe the inhibitory effect of norcantharidin (NCTD) on ovarian cancer 3AO cells and AO cells in vitro, and to explore its mechanism of action. Methods: Cell culture technique was used to determine the inhibitory effect of NCTD on ovarian cancer 3AO and AO cells by crystal violet staining. Flow cytometry was used to analyze the cell cycle changes of two tumor cells under the action of NCTD. RESULTS: NCTD 1 μg/ml had a slight inhibitory effect on 3AO and AO cells, and the IC50 was 20 μg/ml and 10 μg/ml, respectively. The effect of NCTD on 6h, 3AO and AO cell growth was inhibited, and the inhibition rate increased with the prolonged drug action time. Under the action of NCTD, the percentage of cells in GAO and M phase of 3AO and AO cells increased significantly, while the percentage of cells in S phase decreased correspondingly. CONCLUSION: NCTD has significant inhibitory effects on ovarian cancer 3AO and AO cells in vitro, and its inhibition is dose- and time-dependent. The mechanism of NCTD anti-tumor cell proliferation may be related to its specific inhibition of tumor cells in G2+M phase
