目的:采用不同孔径钛铝金属间化合物多孔材料(TAICPM)分离大黄虫丸,探讨经不同孔径TAICPM处理后的大黄虫丸抗动脉血栓形成的作用及机制。方法:选择70只SD雄性大鼠随机分为7组,其中正常组、模型组、对照组各10只,TAICPM组40只:1-5μm组、5-10μm组、10-15μm组、15-20μm组,每组各10只。血栓干重、血小板计数、血栓素B2(TXB-2)和6酮前列腺素F1α(6-keto-PGF1α)作为观察指标,评价经TAICPM分离后大黄虫丸抗动脉血栓形成的效果。结果:①对照组及TAICPM组血栓干重、血小板计数明显少于模型组(P<0.05),TXB-2和6-keto-PGF1α也较模型组有显著性差异(P<0.05);②TAICPM组血小板计数明显低于对照组(P<0.01),TXB-2和6-keto-PGF1α也较对照组有统计学差异(P<0.01);③1-5μm多孔材料组血栓干重明显低于其它3个多孔材料组(P<0.01),血小板计数、TXB-2和6-keto-PGF1α也较其它3组有显著性差异(P<0.01)。结论:经TAICPM处理后的大黄虫丸在抑制动脉血栓形成方面优于未处理的大黄虫丸,其中孔径为1-5μm的多孔材料效果更优。
OBJECTIVE: To isolate Rhubarb worm pills by using different pore sizes of titanium-aluminum intermetallic compound porous material (TAICPM) to investigate the anti-arterial thrombosis effect and mechanism of Rheum officinale pills treated by TAICPM with different diameters. Methods: Seventy male Sprague-Dawley rats were randomly divided into 7 groups: 10 in normal group, model group and control group, 40 in TAICPM group: 1-5μm group, 5-10μm group, 10-15μm group, 15- 20μm group, 10 in each group. Thrombus dry weight, platelet count, TXB-2 and 6-keto-PGF1α were used as indexes to evaluate the anti-arterial thrombosis effect of Dahuangzhewan after TAICPM separation. Results: ① Thrombus dry weight and platelet count in control group and TAICPM group were significantly less than those in model group (P <0.05), TXB-2 and 6-keto-PGF1α were also significantly different from those in model group (P <0.05) (P <0.01). The TXB-2 and 6-keto-PGF1α also had statistical difference compared with the control group (P <0.01). ③ The thrombosis dry weight of 1-5μm porous material group was significantly lower than the other three groups Porous materials group (P <0.01), platelet count, TXB-2 and 6-keto-PGF1α also had significant difference compared with the other three groups (P <0.01). Conclusion: The rhubarb worm treated with TAICPM is better than untreated rhubarb worm in inhibiting arterial thrombosis, and the porous materials with the pore size of 1-5 μm are more effective.
