应用Super Pac C18柱(125mm×4.0mm,3μm),建立了PTG-氨基酸的HPLC分析方法。衍生步骤快速、灵敏、简便。能有效地分析蛋白水解物中的各种一级、二级氨基酸。色谱系统检测灵敏(灵敏度可达40pmole以下),分析快速(18min)。方法线性响应、重复性均佳。应用此法首次确定了新发现的钙结合蛋白兔阑尾GaBP_(21)的氨基酸组份,证明它是一种与CaM相比组成不同、特征相似的钙结合蛋白。
Super Pac C18 column (125mm×4.0mm, 3μm) was used to establish the HPLC method for the determination of PTG-amino acids. Derivative steps are quick, sensitive, and simple. It can effectively analyze various primary and secondary amino acids in protein hydrolysates. The chromatographic system is sensitive (sensitivity up to 40 pmoles) and fast (18 min). The method has good linear response and repeatability. Using this method, the newly identified amino acid component of the newly identified calcium binding protein rabbit tail GaBP_(21) was identified, demonstrating that it is a calcium binding protein with a similar composition and similar characteristics to CaM.
