目的研究老年痴呆(AD)小鼠模型脑神经组织生化工程指标的改变及乌圆补血口服液对其重建的影响。方法将小鼠分成模型组、治疗组和正常组,模型组和治疗组小鼠腹腔注射D-半乳糖和亚硝酸钠混合液,每天1次,连续60 d造模。造模30 d后,治疗组用乌圆补血口服液灌胃治疗,模型组和正常组用等体积蒸馏水灌胃。y型水迷宫测定小鼠治疗前后记忆功能。实验结束,处死小鼠,取大脑制备10%脑匀浆,测定脑匀浆生化指标。结果脑匀浆乙酰胆碱酯酶(TchE)活力模型组、正常组和治疗组分别为(1.709±0.498)IU/mg、(1.436±0.313)IU/mg、(1.182±0.157)IU/mg,模型组明显高于其他2组(P<0.05和P<0.01);丙二醛(MDA)含量分别为(7.935±0.775)μmol/L、(6.934±1.284)μmol/L、(6.356±0.799)μmol/L,模型组明显高于其他2组(P<0.05);脑(O-.2)氧自由基清除率分别为(2.346±1.167)%、(7.366±3.413)%、(8.672±3.519)%,治疗组和正常组均明显高于模型组(P均<0.01);实验前各组水迷宫错误率分别是9.91%,7.41%,14.81%;实验结束时错误率分别是10.10%,1.23%,1.39%,实验结束时模型组水迷宫错误率明显高于治疗组和正常组;其他生化指标脑蛋白含量、脑GSH-PX等各组间无显著性差异。结论 D-半乳糖和亚硝酸钠联合造痴呆模型小鼠大脑神经组织胆碱酯酶活性等生化指标有明显改变,乌圆补血口服液对其重建有明显的促进作用。
Objective To study the changes of biochemical engineering indexes in brain tissue of senile dementia (AD) mice and the effects of Wuyuan Buxue Oral Liquid on their reconstruction. Methods Mice were divided into model group, treatment group and normal group. Mice in model group and treatment group were injected intraperitoneally with D-galactose and sodium nitrite once a day for 60 days. After 30 days of modeling, the treatment group was treated with Wuyuan Buxue Oral Liquid, and the model group and the normal group were given the same volume of distilled water. y-type water maze determination of memory function of mice before and after treatment. At the end of the experiment, the mice were sacrificed and 10% brain homogenate was prepared from the brain to determine the biochemical indicators of brain homogenate. Results The levels of acetylcholinesterase (TchE) in brain homogenate of model group, normal group and treatment group were (1.709 ± 0.498) IU / mg, (1.436 ± 0.313) IU / mg and (6.934 ± 1.284) μmol / L and (6.356 ± 0.799) μmol / L, respectively, which were significantly higher than those in other two groups (P <0.05 and P <0.01) L, the model group was significantly higher than the other two groups (P <0.05); the oxygen free radical scavenging rate of brain (O-2) were (2.346 ± 1.167)%, (7.366 ± 3.413)% and (8.672 ± 3.519) , The treatment group and the normal group were significantly higher than the model group (P <0.01). Before the experiment, the water maze error rates were 9.91%, 7.41% and 14.81% respectively. The error rates at the end of the experiment were 10.10% and 1.23% , 1.39%. At the end of the experiment, the water maze error rate in the model group was significantly higher than that in the treatment group and the normal group. There was no significant difference in brain biochemical parameters such as brain protein content and brain GSH-PX between the two groups. Conclusion D-galactose and sodium nitrite combined with dementia model mice brain tissue of cholinesterase activity and other biochemical indicators have changed significantly, Wuxu Buxue oral liquid to its reconstruction had a significant role in promoting.
