采用31P-核磁共振技术(31P-NMR)建立了无损伤测定水稻根系细胞内pH值的方法。通过测定水稻根系细胞内磷酸盐化学位移间接确定细胞液泡及细胞质内pH,计算得到正常水培条件下水稻根系细胞液泡及细胞质内pH分别为5.43±0.05和7.45±0.04。另外,用该方法还测定了磷饥饿条件下水稻根细胞内pH的变化。
The method of 31P-NMR (31P-NMR) was established to determine the intracellular pH of rice roots without damage. The vacuolar and cytoplasmic pH of rice root cells under normal hydroponic culture were calculated to be 5.43 ± 0.05 and 7.45 ± 0.04, respectively, by determining the intracellular vacuolar and cytoplasmic pH indirectly by measuring the chemical shifts of phosphate in rice roots. In addition, the method was also used to determine the change of pH in rice root cells under phosphorus starvation.
