目的:研究薯蓣皂苷对正常大鼠心室肌细胞L-钙电流(ICa,L)的影响,旨在探讨薯蓣皂苷对心肌缺血再灌注(I/R)损伤保护作用的可能机制。方法:酶解法分离大鼠单个心室肌细胞,应用全细胞膜片钳技术记录薯蓣皂苷低(10μg/ml)、中(20μg/ml)、高(30μg/ml)三个剂量对大鼠单个心室肌细胞ICa,L的影响。结果:薯蓣皂苷10μg/ml不影响ICa,L峰值,而20μg/ml和30μg/ml能使ICa,L峰值降低,抑制呈剂量依赖性;20μg/ml和30μg/ml薯蓣皂苷使I-V曲线上移,内向电流减小,但基本不改变曲线的形状。30μg/ml薯蓣皂苷使失活曲线向负电位方向变化,曲线左移,各剂量均对激活曲线无影响。结论:薯蓣皂苷通过促进ICa,L失活的过程抑制正常大鼠心室肌细胞I,减小I峰值,这可能是其抗缺血再灌所致心律失常的作用机制之一。
Aims: To study the effects of diosgenin on L-type calcium currents (ICa, L) in normal rat ventricular myocytes and to explore the possible mechanism of diosgenin in the protective effect of myocardial ischemia-reperfusion (I / R) injury. CONCLUSION: Diosgenin inhibits I and decreases I peak in normal rat ventricular myocytes by promoting the inactivation of ICa and L, which may be one of the mechanisms of anti-ischemia reperfusion-induced arrhythmia.
