应用人白细胞抗原(HLA)基因的聚合酶链反应-限制性片段长度多态性核苷酸分型技术,以等位基因特异性的限制性内切酶(ApalⅠ、 BsajⅠ、 HphⅠ、 FokⅠ、 Mbo Ⅱ、 MulⅠ)消化 DQA1座位特异的 PCR扩增产物,对50例胃溃疡患者、 50例正常人的HLA-DQA1等位基因进行分析,旨在研究武汉地区汉族人HLA-DQA1基因与胃溃疡的相互关系。发现胃溃疡患者DQA1*0301显著增加(P<0.05),而DQA1*0102显著下降(P<0.05)。提示*QA1*0301和*QA1*0102与胃溃疡有一定关联,胃溃疡患者与正常人之间存在着免疫遗传异质性的养异。
The allele-specific restriction endonucleases (ApalⅠ, BsajⅠ, HphⅠ, FokⅠ, Mbo) were cloned by polymerase chain reaction-restriction fragment length polymorphism (PCR) using human leukocyte antigen (HLA) Ⅱ, Mul Ⅰ) to digest DQA1 locus-specific PCR amplification products. HLA-DQA1 alleles in 50 patients with gastric ulcer and 50 normal individuals were analyzed to investigate the relationship between HLA-DQA1 gene and gastric ulcer Interrelationship. DQA1 * 0301 was found to be significantly increased in patients with gastric ulcer (P <0.05), while DQA1 * 0102 was significantly decreased (P <0.05). Prompt * QA1 * 0301 and * QA1 * 0102 and gastric ulcer have a certain correlation, gastric ulcer patients and normal there is a heterogeneous immune genetic heterogeneity.
