目的 :观察体外培养的牙周组织三种主要功能细胞的生物学特性差异。方法 :对人牙龈成纤维细胞 (GFs)、牙周韧带细胞 (PDLCs)及牙槽骨细胞 (ABCs)进行分离、培养、传代后 ,用MTT比色法、碱性磷酸酶测定法、考马斯亮蓝法及茜素红染色法测定其生物学活性。结果 :PDLCs和ABCs在碱性磷酸酶活性 (ALP)及矿化结节形成方面与GFs有显著性差异。结论 :尽管三种细胞具有相同的镜下表现 ,但生物学特性不同 ,则具有不同的表型 ,从而在牙周再生中发挥着不同的作用。
OBJECTIVE: To observe the difference of biological characteristics of three major functional cells cultured in vitro. Methods: Human gingival fibroblasts (GFs), periodontal ligament cells (PDLCs) and alveolar bone cells (ABCs) were isolated, cultured and passaged. MTT colorimetric assay, alkaline phosphatase assay, The biological activity was determined by blue method and alizarin red staining. RESULTS: PDLCs and ABCs had significant differences from GFs in the formation of alkaline phosphatase (ALP) and mineralized nodules. CONCLUSION: Although all three cells have the same microscopic appearance but different biological characteristics, they have different phenotypes and thus play different roles in periodontal regeneration.
